Objective:To investigate the relationship between C677T MTHFR Hcy and its metabolic key enzyme gene polymorphism and OSAHS related hypertension.Methods:1.Research objects and groupsA total of 135 cases of continuous subjects were collected in Kunming Yanan Hospital and divided into 3 groups:1.There were a total of 30 cases in the health control group,of which 14 were men and 16 were women,and the average age was 51.93 ± 6.88 years old.2.A total of 35 cases of high blood pressure(35 cases),15 cases of men and 20 cases of women,with an average age of 52.31 ± 7.23 years;3.Hypertension combined with OSAHS group(70 cases),33 men and 37 women,with an average age of 54.51 ± 6.24 years.2.Determination of serum homocysteine(Hcy):The 2ml venous blood of subjects was taken in the early morning and to be sent to our hospital clinical laboratory to use immuno turbidimetryto detect serum homocysteine(homocysteine,Hcy)level.3.MTHFR detectionThe venous blood was collected from the study subjects.After the extraction of whole blood genome DNA and the amplification of the PCR gene,the PCR amplification products were obtained.The genotype of MTHFR C677T site was sequenced by ABI 3730XL DNA sequencing.The base site of MTHFR gene is C,and the base of mutation site is T.Results:1.Compared with the healthy control serum Hcy level,the serum level of Hcy in the simple hypertension group was significantly increased(7.92±1.94umol/L,10.54±2.01 umol/L,P<0.05),and the level of Hcy in the hypertension combined with OSAHS group was significantly increased(7.92±1.94umol/L,15.19±5.66 umol/L,P<0.05).The level of serum Hcy in patients with hypertension combined with OSAHS was significantly higher than that of serum Hcy in simple hypertension group(15.19±5.66umol/L,10.54±2.01 umol]L,P<0.05).2.In a total of 30 cases in healthy controls,the proportion of MTHFR C677T loci CC,CT and TT genotypes were 60%,16.7%,23.3%,35 cases in simple hypertension group were 42.9%,40%,17.1%,70 cases in hypertension combined with OSAHS were 12.9%,57.1%,30%.There was no significant difference in genotype distribution between healthy controls and simple hypertension group(P>0.05).There was a significant difference in genotype distribution between the hypertension combined with OSAHS group and the simple hypertension group(P<0.05).There was a significant difference in the distribution of genotypes between the hypertension combined with OSAHS group and healthy controls(P<0.05).3.The serum Hcy levels of CC genotype,CT genotype and TT genotype were 9.71±2.66umol/L,13.11±5.41umol/L and 14.36±6.21umol/L respectively.Compared with the level of serum Hcy in CC genotypes,the level of serum Hcy in CT genotype was significantly higher(9.71 ±2.66umol/L,13.11 ±5.41umol/L,P<0.05),and the level of serum Hcy in TT genotypes was significantly higher(9.71±2.66umol/L,14.36±6.21umol/L,P<0.05).There was no significant difference in serum Hcy level between the TT genotype and the serum Hcy level of CT genotype(14.36±6.21umol/L,13.11±5.41umol/L,P>0.05).Conclusions:1.Compared with the healthy control group,the serum homocysteine level was significantly higher in the simple hypertension group and the OSAHS-related hypertension group.2.There was significant difference in genotype distribution of MTHFR C677T locus in hypertension and hypertension combined with OSAHS group compared with healthy control group and simple hypertension group.3.The levels of serum homocysteine in CT and TT genotypes of MTHFR C677T locus were significantly higher than those in CC genotypes.
