Objective: To investigate the gene polymorphism and gene expression of TOLL-like receptor 4(TLR4)gene(at the gene locus of rs56101219 and rs5030722)in peripheral blood of patients with sepsis,and to explore its relationship with the expression of inflammatory factors TREM-1,1L-6,and 1L-2 MCP-1 and the correlation with sepsis.Methods: The genomic DNA was obtained by venous blood collection from 24 normal individuals and 24 sepsis patients.The TLR4 gene(rs56101219,rs5030722)was cloned by polymerase chain reaction and sequenced to analyze the gene polymorphism.Peripheral blood mononuclear cells were extracted and the expression of TLR4 mRNA was detected by real-time fluorescence quantitative PCR.The expression of inflammatory factors such as TREM-1,IL-6,IL-2 and MCP-1 was detected by enzyme-linked immunosorbent assay.ROC curve was used to analyze the correlation between TLR4 and TREM-1,IL-6,IL-2 and MCP-1,and to explore its predictive value for diagnosis of sepsis.Results: No polymorphism was found in the gene locus of rs56101219 andrs5030722 at TLR4 gene.The expression of TLR4 mRNA in the peripheral blood of patients with sepsis was higher than that in the healthy group.The expression of inflammatory cytokines TREM-1,IL-6,IL-2,MCP-1 in sepsis was markedly increased.And the correlation with TLR4 mRNA was significant.Conclusion: There is no gene polymorphism was found in the gene locus of rs56101219 and rs5030722 at TOLL-like receptor 4(TLR4)gene in peripheral blood of patients with sepsis.TLR4 expression in peripheral blood of patients with sepsis and TREM-1,IL-6,IL-2,MCP-1 and other inflammatory factors have a significant correlation,can serve as an early warning and diagnostic indicators of sepsis.
