Objective: This study took SKVO3 human ovarian cancer cell model of nude mice as the research object to explore the effect and influence of high-frequency bipolar pulse electric field on tumor ablation of nude mice under the skin.Methods: Establishing SKVO3 vaccinated people ovarian cancer cells in nude mice model,provided by the institute of electrical engineering,the pulse generator from Chongqing university,adopts plate electrode method,according to the previous cell screening experiment set categories,respectively pulse width 2?s(50 is the pulse count),5?s(20 is the pulse count),combined pulse(2?s +100?s)and field strength above is 2000 v/cm,pulse sequence number 45.1.Tumor volume changes after treatment with high frequency bipolar pulse electric field.2.Using acceleration sensor to measure muscle contraction degree.3.Testing the number of neutrophils,monocytes and macrophages in peripheral blood of nude mice to understand whether the pulsed electric field has specific inflammatory and anti-tumor mechanisms for nude mice.4.ELISA method was used to detect the non-specific anti-tumor effect of TNF-a and IFN-? in nude mice.5.NK cell activity was detected by spleen cytotoxicity.6.The tumor ablation effect was observed under subcutaneous tumor of nude mice.7.To observe the general conditions of the nude mice,to determine the muscle contraction and tolerance,and to draw the growth curve to determine the impact of pulse electric field on the survival time of the rat.Results: 1.The changes of tumor volume according to the treatment of electric field ablation,5?s complete tumor ablation pulse width experimental nude mice and no recurrence,2?s pulse width group and composite pulse group have different degree of recurrence or invalid.2.Observation of the peak value of waveform under the action of high frequency bipolar pulse electric field showed that the strength of traditional pulse field decreased significantly and the muscle contraction weakened.3.By groups of nude mice peripheral blood cell count,5?s pulse width group and composite pulse width experimental nude mice on the number of total number of cells,neutrophils and monocytes detection ratio increased significantly,5?s group is superior to the combination of pulse group is better than the 2?s pulse width(P<0.05).4.The contents of TNF-a and IFN-? in the serum of the nude mice were detected by ELISA method,and the content of each pulse treatment group was significantly increased,and the increase was the most significant in the 5?s(P<0.05).5.The spleen cells of nude mice were extracted from each group to measure the toxicity of spleen cells.6.By observing the HE staining slices,see 5?s pulse width experimental sample after processing the entire tumor tissue in cytoplasm staining change,cellular structure damage is not clear,karyorrhexis disappear or dissolved,melt completely,the experimental group were residual tumor cells.7.According to the growth curve of mice,each group nude mice survival time a tumor-burdened blank control group was also extended to different extent,of which 5?s pulse width experimental nude mice survival days is greater than the combined pulse.There was no death or intolerance in the field of nude mice(P<0.05).Conclusion: 1.The optimal pulse width and intensity were studied by using the high frequency bipolar pulse electric field splint electrode for the tissue ablation of nude mice.2.High frequency bipolar pulsed electric field is effective for the ablation of SKVO3 cell solid tumor in nude mice,to melt completely,and compared with traditional pulse electric fields of early experiments,experimental animal muscle contraction weakened obviously provides a basis for subsequent physical minimally invasive treatment of ovarian cancer.
