Objective To establish the neonatal rats model of periventricular Leukomalacia(PVL),and test the expression of myelin basic protein(MBP)and the tau protein in the zone of the white matter,combined with the newborn mice growth and pathologic change of brain tissue,evaluate the neuroprotective effect of ginsenoside Rd(GSRd)in the neonatal rats with PVL.Methods Three-old-days newborn SD rats were made by isolating and ligating the left common carotid artery and inhaling mixed gas of 92%N2+8%O2 for 1 hour,the experiment seted up three groups,PVL model group(ligation + hypoxia,given an intraperitoneal injection with saline solution)and the GSRd trial group(ligation + hypoxia,given an intraperitoneal injection with ginsenoside Rd)and the sham opertion group(only separated the left common carotid artery,without ligation and hypoxia,given an intraperitoneal injection of isometric saline solution);8 rats in each groups were randomly sacrificed at the 3th day,7th day and 14 th day after operation.pathological change of brain was observed by HE staining,the expression of MBP,tau protein were measured with immunohistochemistry in the zone of white matter around the lateral cerebral ventricle of three groups,in order to investigated the neuroprotective effect of ginsenoside Rd on the white matter injury of the neonatal rat model of periventricular leukomalacia.Results1.The pathological change of brain with HE in the three groupsThe damage in cerebral cortex of the PVL model group and the GSRd trial group was mild,the lesions mainly occured in the white matter area around the ventricle.The white matter of the GSRd trial group and PVL model group after operation 3th day appeared loose structure,disorder,cell edema,necrosis;at 7th days after operation,the leukoaraiosis edema reduced slightly,hyperplasia of microglia and astrocytes were found,their white matter around the ventricle formed liquefaction necrosis areas and sacs,and their left lateral ventricle was expanded slightly in the PVL model;Fourteen days after the operation,the malacia and liquefaction necrosis in the white matter of the rats were observed,the left ventricle is obviously enlarged than right ventricle.the pathological changes in the PVL model group was most serious,the pathological injury of the GS-Rd trial group was mild than the PVL model group;and there was no pathological change in the sham operation group.2.Immunohistochemistry staining and image quantitative analysis2.1 The expression of MBP in white matter around lateral cerebral ventricle The expression of MBP showed a rising trend with the increase of age in the three groups,At the same time-point,sham operation group was maximum closely followed by the GSRd trial group,PVL model group was minimum,the differences between groups were statistically significant(P < 0.05).2.2 The expression of tau in white matter around lateral cerebral ventricle Accompany with the increase of age of rats,the expression of tau protein of white matter around the ventricle was raise in three groups,At the same time-point,sham operation group was maximum,closely followed by the GSRd trial group,the PVL model group was minimum,the differences between groups were statistically significant(P < 0.05).Conclusions:1.Ginsenoside Rd can relieve the structural loosening,cell edema,necrosis and malacoma of white matter in PVL model rats,and lessen the extent of ventricular enlargement.It suggested that ginsenosides have neuroprotective effects for the white matter injury of the neonatal rat model of periventricular leukomalacia2.Ginsenoside Rd can increase the expression of MBP,and reduce the expression level of tau protein in the periventricular white matter of the neonatal rat with PVL,that means ginsenoside Rd involved in promoting oligodendrocyte differentiation and maturation,decreasing axonal disorder,improved myelination,it suggested that ginsenoside Rd has neuroprotective effect on the neonatal rat model of periventricular leukomalacia.
