Objective: This study used proteomics technology of dimensional fluorescent difference in gel electrophoresis(2 D-DIGE)technology combined matrix-assisted laser desorption time of flight mass spectrometry(MALDI-TOF-MS)to screen the proteins differential expression in proteinuria?+ and proteinuria?3+ of spontaneous NZB/W F1 mice of systemic lupus erythematosus(SLE),providing new clues of the diagnosis and monitoring of the disease activity of lupus nephritis from perspective of proteomics.Methods: Firstly,with the glomerular specimens of spontaneous NZB/W F1 mice of SLE as the research object,and the glomerular specimens of 28 weeks of C57 BL / 6 as the control group,adopting the method of kidney filled of magnetic beads to getting glomerular tissue in mice,divided into 1)proteinuria protein?+ group,2)proteinuria protein?3+ group,3)28 weeks C57 BL / 6 group.Subsequently,performing 2D-DIGE after making each group with the Cy Dye.With the software to select the protein spots of differences at a multiple of 1.4 or more,then performed MALDI-TOF – MS to get the protein expression profile of proteinuria?+ and proteinuria?3+ of spontaneous NZB/W F1 mice of SLE.Finally,western blot and immunohistochemical validation were performed on RRAGB,SMG1,MMSA,ANGP2,ATPB and other proteins.Results: A total of 57 expressed differentially proteins were obtained by mass spectrometry of133 protein spots.Among the 57 proteins,comparing the groups of proteinuria?+ and proteinuria?3+ of NZB/W F1 mice obtained 18 proteins of up-regulated expression and 24 proteins of down-regulated expression;comparing the groups of proteinuria?3+ of NZB/W F1 mice and C57BL/6 of 28 weeks obtained one protein of up-regulated expression and 11 protein of down-regulated expression;comparing the groups of proteinuria?+ of NZB/W F1 mice and C57BL/6 of 28 weeks obtained one protein of down-regulated expression.Conclusions : 57 expressed differentially proteins were selected by the proteomics study of protein of glomerular of NZB/W F1 mice.On the basis of the difference multiple in protein?protein matching of mass spectrometry ? analysis of function and LN correlation,and verification of Western blot and immunohistochemistry,RRAGB,SMG1,MMSA,ANGP2,ATPB may be the target proteins,providing new clues of the diagnosis and monitoring of the disease activity of lupus nephritis from perspective of proteomics.
