Objective:Several cytokines play an important role in the development of SLE.The objective of this study is to detect serum levels of BlyS in patients with SLE,other connective tissue diseases without SLE and normal controls,in patients with or without Lupus nephritis and in before and after therapy of SLE patients.Analyze the relationships between serum levels of BlyS and IgG,IgA,IgM,complement C3,C4,antibody ds-DNA,24-hour urine proteinuria,SLEDAI degree,ESR and C reaction protein.To explore the possible roles of BlyS in the pathogenesis of SLE, especially with renal injury.Try to find a special and sensitive biologic factor to help the diagnosis of SLE,to predict the prognosis and to evaluate the therapy effection. Try to provide clues to reveal the pathogenesis of SLE.Methods:The serum levels of BlyS were assayed in 30 inpatients with SLE during Mar 2007 to Oct 2007,20 patients with other connective tissue diseases and 20 healthy controls by ELISA.Analyze different serum levels of BlyS in patients with SLE,other connective tissue diseases without SLE and normal controls,in patients with or without Lupus nephritis and in before and after therapy of SLE patients. Analyze the relationships between serum levels of BlyS and IgG,IgA,IgM,complement C3,C4,antibody ds-DNA,24-hour urine proteinuria,SLEDAI degree,ESR and C reaction protein.Results:1.The serum level of BlyS was significantly higher in SLE groups than other connective tissue disease groups and normal control groups,P<0.05;the serum levels of BlyS were significantly higher in other connective tissue disease groups than normal control groups,P<0.05。2.The serum level of BlyS was significantly higher in patients with active groups than inactive SLE groups and normal control groups,P<0.05;the serum levels of BlyS were significantly higher in inactive SLE groups than normal control groups, P<0.05。3.The serum level of BlyS was significantly higher in patients with Lupus nephritis than patients without Lupus nephritis groups and normal control groups,P<0.05;the serum levels of BlyS were significantly higher in SLE patients without Lupus nephritis than normal control groups,P<0.05。4.The serum level of BlyS was significantly higher in SLE patients before therapy than after therapy,P<0.05。5.The serum level of BlyS has positive correlation with the serum levels of IgG,IgA,IgM,antibody ds-DNA,24-hour urine proteinuria,C reaction protein and SLEDAI,P<0.05;the serum levels of BlyS have negative correlation with the levels of complement C3,P<0.05;and the serum levels of BlyS have no correlation with the levels of complement C4,ESR and the duration of the disease,P>0.05。Conclusions:1.The serum level of BlyS was significantly increased in SLE patients depending on the disease activity and correlated with the SLEDAI score.The measurement of serum GM-CSF and GM-CSF Ab may be two useful indicators in SLE disease activity.2.The serum level of BlyS may predict renal injury in SLE.The measurement of serum BlyS may be a useful indicator in SLE with renal involvement.The serum level of BlyS combinations with the clinical symptoms and the indicators of laboratory examination in patients with SLE maybe the useful noninvase tool for monitoring the Lupus nephritis activity.3.The serum level of BlyS was significantly descended after treatment in SLE patients,so serum level of BlyS may give some directions to SLE prognosis,and may evaluate the therapy effection.4.The serum level of BlyS may be one of the diagnose index of SLE,and can determine disease activity.BlyS may be the drug targets of biotherapy in patients with SLE.
