OBJECTIVE: To detect the expression of CD14 and CD16 on decidual macrophages in pregnant women with severe preeclampsia and normal pregnant women,and to analyze its role in the pathogenesis of preeclampsia.Methods: From June 2017 to December 2017,the cesarean delivery in Shengjing hospital was divided into two groups: the control group and the case group.There was no significant difference in the age and gestational age between the two groups,No previous hypertension,diabetes,chronic nephritis,hepatitis,hyperthyroidism,infection and other diseases,no pregnancy complications,not taking immunosuppression or enhanced drugs,are singleton pregnancies.The case group was severe preeclampsia(based on the diagnosis and classification criteria of the eighth edition of Obstetrics and Gynecology)edited by Xie Xing.There were 15 patients(aged 34.46±6.88 years,gestational age 36.18±1.18 weeks).The control group of 15 normal pregnant women,age 33.33 ± 5.48 years,gestational age was 37.12±0.88 weeks.There was no significant difference in gestational age and age between the two groups by statistical test.Elective elective cesarean section surgery,the oval clamp take the placenta attached to the uterine decidua 5g.Fresh decidua removed blood clots,scissors cut with ophthalmology,lymphocyte separation fluid extracted mononuclear cells,set up two test tubes,experimental tube and negative control tube.Antibody CD14-FITC and CD16-PE were added into the experimental tube in sequence of 10?1.Antibody CD14-FITC and 10?1 of CD16-PE were added to the control tube,and then 50?1 of the extracted mononuclear cell suspension was added to each tube.After the sample is added,the turbulent mix,avoid light for 30 minutes.Each tube was then added with 400?1 PBS and resuspended.The expression of CD14 and CD16 on CD14-FITC and CD16-PE macrophages was detected by flow cytometry.The experimental data using SPSS 23.0 statistical package on the measurement data t test.P <0.05 for the difference was statistically significant.Results: The expression of CD14 + in decidual macrophages in severe preeclampsia group was 36.26%±5.66%,which was significantly higher than that in normal pregnancy group23.33%±4.95% t = 8.493,P<0.05.The expression of CD14 + CD16 + on decidual macrophages in severe preeclampsia group was 8.98%±3.87%,slightly higher than that in normal pregnancy group(7.99%±2.15%,t =-1.931,P>0.05)Significance of learning.Conclusion: The results of this study showed that the levels of CD14 + CD16-in decidual macrophages in patients with severe preeclampsia were slightly higher than those in normal pregnant women,indicating that the macrophages in the decidua of preeclampsia mainly come from The expression of CD14 + CD16 + in decidual tissue of peripheral blood mononuclear cells and severe preeclampsia patients was higher than that of normal pregnancy group,indicating that decidual macrophages may participate in the pathogenesis of preeclampsia process.
