Objective:Cervical cancer is the most common female reproductive system malignant tumor that occurs in the vaginal area of the cervix.The development of normal cervical epithelium for cervical cancer requires longer precancerous lesions.This stage involves multiple processes and multiple factors,and its mechanism has not yet been clarified.More than 99.7%of cervical cancer can detect high-risk HPV(high-papilloma virus,HR-HPV)[1],but HR-HPV infection is a result of cervical cancer Of the necessary inadequate conditions,in the diagnosis of cervical cancer should also consider other factors.Inactivation of tumor suppressor genes and the activation of related signaling pathways also promote the carcinogenesis of the cervix.G protein-coupled receptor 30(G protein-coupled receptor 30,GPR30)is a novel estrogen receptor.The study found that it is related to the occurrence and development of breast cancer,endometrial cancer,and ovarian cancer,but about GPR30 is rarely studied in the development of cervical cancer.P53 gene(Tumor Protein 53)is an important tumor suppressor gene[2],divided into wild-type and mutant.Mutations in P53 gene lose the function of inhibiting tumorigenesis.Mutant P53 can promote tumor development and is usually highly expressed in tumor tissues.This issue uses immunohistochemistry to detect chronic cervicitis,low-grade squamous intraepithelial lesion(LSIL)and high-grade squamous intraepithelial lesion In the expression of GPR30,P53protein in cervical squamous intraepithelial lesions in different levels of development and analysis of the relationship between the role and the relationship between the diagnosis and treatment of cervical squamous intraepithelial lesions provide a new idea for the diagnosis and treatment of Secondary prevention of cervical cancer provide the theoretical basis.Method:1 This study selected from May 2016 to September 2017 due to cervical lesions in Hebei Provincial People's Hospital gynecology clinic,cervical circumcision(LEEP)treatment of a total of 65 cases,including 25 cases of LSIL,HSIL 40 cases,At the same time select the same period due to benign uterine hysterectomy in patients with pathological cervix for chronic cervicitis in 20 cases as a control group.2 The expression of GPR30 and P53 in three groups were detected by SP method,and then the correlation between GPR30 and P53 protein was analyzed.3 Using SPSS21.0 statistical software for statistical analysis,?=0.05 for the test level,when P<0.05 can be considered statistically significant differences.4 Spearman correlation test to analyze the relationship between the two expression in cervical squamous intraepithelial lesion.Result:1 GPR30 protein expression in each groupThe positive rates of GPR30 protein in chronic cervicitis,LSIL and HSIL tissues were 20.00%(4/20),52.00%(13/25)and 87.5%(35/40),respectively.There was significant difference between the three groups Statistical significance(?~2=26.83,P<0.05),Chi-square split between two pairs of comparative differences were statistically significant(?~2=4.84,P<0.05;?~2=13.37,P<0.05;?~2=30.93,P<0.05).(Table 1 and Fig.1)2 P53protein expression in each groupThe positive expression rates of P53 protein in chronic cervicitis group,LSIL group and HSIL group were 0.00%(0/20),40.00%(10/25)and 75.00%(30/40),respectively(?~2=10.29,P<0.05;?~2=7.96,P<0.05;?~2=30.33,P<0.05;),and the differences between the two groups were statistically significant<0.05).(See Table 2 and Fig.2)3 In cervical squamous intraepithelial lesions,there was a positive correlation between GPR30 protein and P53 protein expression.The Spearman rank correlation coefficient was r=0.727,P<0.05.(Table 3)Conclusion:1 The positive rate of GPR30 and P53 protein in LSIL and HSIL tissues was higher,and increased with the increase of cervical lesion grade.Both of them may play a catalytic role in the occurrence and development of cervical squamous intraepithelial lesions.2 GPR30 protein and P53 protein play a synergistic role in the occurrence and development of cervical squamous intraepithelial lesions.
