Objective:The normal values of insulin-like growth factor-1 and insulin-like growth factor binding protein-3 in healthy children aged 7-18 years in Chongqing were established by testing blood samples from normal children in Chongqing. And explore their relationship with age, gender, height, weight, and stage of puberty development. Methods:A total of 1024 healthy children aged 7-18 years(477 males and 547 females)from 6 primary and secondary schools in Chongqing were selected and their age, sex, height, weight, and stage of puberty development were measured and recorded.Fasting serum IGF-1 and IGFBP-3 levels were measured by chemiluminescence. The relationship between IGF-1,IGFBP-3 and age, sex, height, weight, and stage of puberty development were studied by t test and multiple regression analysis.Results:1.The peak serum IGF-1 levels in boys and girls occurred in the 13 to 15 year old group and the 11 to 13 year old group, respectively.Compared with boys and girls of the same age group, serum IGF-1 levelsof boys aged 7 to 13 and 17 were lower than girls.IGF-1 levels in boys aged 14 to 16 and 18 were higher than girls(7 to 12 years old group P<0.05,the difference was statistically significant.13 to 18 years old group P>0.05,the difference was not statistically significant).2.The peak of serum IGFBP-3 in boys and girls appeared in the 14 to16 year old group and 13 to 15 year old group, respectively.Compared with boys and girls in the same age group, the serum levels of IGFBP-3 in boys aged 7 to 13 were lower than girls, and the serum levels of IGFBP-3 in boys aged 14 to 18 were higher than girls(8 and 10 year old group P <0.05,the difference was statistically significant. The other groups P>0.05,the difference was not statistically significant).3.The serum IGF-1 values of boys and girls peaked at stage IV and III respectively.Compared with boys and girls at the same stage of development,IGF-1 serum levels of I-III boys were lower than girls, and IV-V boys serum IGF-1 levels were higher than girls(I-III period P <0.05,the difference was statistically significant.IV-V period P>0.05,the difference was not statistically significant).4.The serum IGFBP-3 values of boys and girls peaked at V and III stages, respectively.Compared with boys and girls at the same stage of development, serum IGFBP-3 levels in boys I to III were lower than girls, and IGFBP-3 levels in boys IV to V were higher than girls(I,IV ~ V period P <0.05,the difference was statistically significant.Stage II,III P>0.05,thedifference was not statistically significant.).5.Using IGF-1 as the dependent variable, the ? values of age and height were the highest, which were-0.49 and 0.596 respectively. The gender, developmental stage, and body weight were approximately equal to each other and were all relatively small(P<0.05).Using IGFBP-3 as a dependent variable, the ? values of gender, age, developmental stage, height, and weight were approximately equal and both were small(P>0.05).Conclusion:1.From school age to puberty, the levels of serum IGF-1and IGFBP-3 in boys and girls showed a first increase with the growth of the age or stage of pubertal development, reached the plateau after reaching the peak, and then slightly decreased and remained at a position slightly lower than the plateau position(Boys' serum IGFBP-3 levels only increased with the increase of stage of pubertal development and entered the plateau period without decreasing).2.The peak serum IGF-1 values of boys and girls occurred in the 13 to15 year group/IV period and 11 to 13 year group/III period respectively. The peak of IGFBP-3 values appeared in the 14 to 16 year group/V period,13 to 15 year group / III period.3.In the same age group/stage of puberty development, serum IGF-1and IGFBP-3 were different between boys and girls, and the girl entered the peak stage of IGF-1 and IGFBP-3 about 1 to 2 years of age/1 to 2stages of pubertal development in advance of the boy.4.Height and age have a greater impact on IGF-1 values.Gender, developmental stage, and body weight have less influence on IGF-1values.The effects of gender, age, developmental stage, height, and body weight on IGFBP-3 values were not statistically significant.5.The results of this survey are similar to those of previous surveys but they are also different. Therefore,it is recommended that all regions conduct investigations and studies to establish normal reference values for IGF-1 and IGFBP-3 in children in their own region.
