Objective To investigate relationship between the expression of estrogen receptor alpha(ER?)in placenta and its gene polymorphism with gestational diabetes mellitus(GDM)in the Han people from North China.Methods 286 patients with GDM and 325 control subjects were collected in the study.ER?genotypes were detected by Polymerase chain reaction-based restriction fragment length polymorphisms(PCR-RFLP)method.The expression of ER?in placentas from 42GDM patients and 51 normal pregnant women were detected by Western blot.Results(1)The frequencies of CC and TT genotype of PvuII in ER?were higher in the patients with GDM than that in controls(c~2=5.536,odds ratio[0R],1.480;confidence interval[CI],0.86 to 3.28;P<0.05).C levels of GDM group were higher than that in control group(c2=5.086;0R,1.372;CI,1.042 to 1.806,P<0.05).(2)Frequencies of CC and CT genotypes of PvuII in ER?gene had higher FPG,FIN values and HOMA-IR compared with those with the TT genotype among patients with GDM(P<0.05).(3)The expressions of ER?were significantly higher in GDM group than that in control group(1.23±0.48vs1.04±0.39;t=2.11).(4)The expressions of ER?were significantly higher in the patients with CC and CT genotype(1.31±0.74)than patients with TT genotype(0.92±0.52,P<0.05).Conclusion The polymorphisms of PvuII in ER?and its high expression in the placenta may be one of the molecular mechanisms leading to insulin resistance in patients with GDM.
