| Objective: To observe the expression and changes of Notch1,HERP1 and bFGF in rabbits with SANFH before and after modeling and after modeling.The pathological changes of rabbit femoral head bone tissue specimens were detected by light and electron microscopy,so as to observe the efficacy of Huoxue Tongluo Decoction in preventing and treating SANFH and to explore its possible mechanism.Methods: Methods 192 rabbits were randomly divided into A,B,C and D groups(n= 48)in each group(n=four).All rabbits were fed for one week.A,B,C three groups in third weeks began to make the mold,the mold cycle is 6 weeks.A group in second weeks began to feed Chinese herbal feed.B group in third weeks began to feed Chinese herbal feed.C,D two groups were fed the same amount of ordinary feed.The gross morphology of the rabbits in each group was observed dynamically,and the bone tissue specimens of the femoral head were observed under the microscope at the end of the second,fifth,eighth week,and calculate the ratio of empty lacuna.The content of bFGF in the blood of rabbits was detected by Elisa method.The bFGF optical density of rabbit bone tissue was detected by S-P(streptavidin-perosidase),Streptomyces avidin peroxidase linked method.Results:1.The ratio of empty lacuna: Each time node,Group C was significantly higher than group D(P<0.01),A and B group were significantly lower than C group(P<0.01);group comparison,C group increased the most,followed by B group,A and D group increased the smallest.2.Notch1: At the end of second weekend,A,B and C groups were significantly higher than those of D group(P<0.01).At the end of fifth weeks,there was no significant difference between the groups(P>0.05);at the eighth weekend,A and B group were significantly lower than those of C group(P<0.01),but there was no significant difference between A group and C group.Within group comparison,A group eighth weeks than second weeks Shaogao,B group for eighth weeks and slightly less than second weeks,C group and D group increased than A group.3.HERP1: At the end of second weekend,there was no significant difference between the groups(P>0.05).At the fifth weekend,B and C groups were significantly higher than those of D group(P<0.01),A group was lower than that of C group(P<0.05),A and B group had no statistical significance(P>0.05).4.bFGF: At the end of the second week,A group was higher than that of B group and C group(P<0.05),A and B compared with the D group,C group(P>0.05),the difference was not statistically significant;the fifth weekend,the comparison between groups(P>0.05),the difference was not statistically significant;at the end of the eighth week,C group was significantly lower than D group(P<0.01),B group than in D group(P<0.05,A),D group(P>0.05),the difference was not statistically significant.Conclusion:1.Huoxue Tongluo Decoction can decrease the rate of empty lacunae,promote the repair of bone necrosis,ischemic necrosis of the femoral head has remarkable curative effect and preventive effect.2.Huoxue Tongluo Decoction can restrict the expression of Notch1 and HERP1 and increase the level of b FGF expression.3.Through the study of Notch1,HERP1 and b FGF,the possible mechanism of Huoxue Tongluo Decoction in the treatment of SANFH is discussed: 1.Huoxue Tongluo Decoction plays a role in the treatment of SANFH by regulating the expression of Notch1 and HERP1 and regulating the proliferation,differentiation or apoptosis of the related cells.2,Huoxue Tongluo Decoction by up regulating the expression of b FGF,repair the related blood vessels,from the recovery of blood supply to play the role of the treatment of SANFH.3,Huoxue Tongluo Decoction can increase the expression of b FGF,promote the synthesis of osteoblasts,inhibit the formation of osteoclasts,and prevent the apoptosis of chondrocytes.4,under the effect of Huoxue Tongluo Decoction can activate a pathway of the Notch pathway,such as HERP1,Notch1-b FGF or Notch1-Notch1-to activate the HERP1-b FGF pathway,and increase the expression of intracellular NICD,which involved in cell proliferation and differentiation and apoptosis,and promote osteogenic differentiation or reduce osteoclast differentiation. |
