Objective: To observe the expression and changes of Notch3,BMP-2 and TGF-β in rabbits with SANFH before and after modeling and after modeling.The pathological changes of rabbit femoral head were observed by microscope,and the effect and mechanism of Huoxue Tongluo Decoction on prevention and treatment of SANFH were discussed.Methods: Methods 192 rabbits were randomly divided into A,B,C and D groups(n= 48)in each group(n=four).All rabbits were fed for one week.A,B,C three groups in third weeks began to make the mold,the mold cycle is 6 weeks.A group in second weeks began to feed Chinese herbal feed.B group in third weeks began to feed Chinese herbal feed.C,D two groups were fed the same amount of ordinary feed.The gross morphology of rabbits in each group was observed dynamically,RT-PCR was used to determine the content of Notch3 in rabbit serum before and after modeling,Elisa was used to detect the content of BMP-2 and TGF-β in rabbit blood.The interception of bilateral femoral headspecimens for pathological examination,and calculate the ratio of empty lacuna.Results: 1.Notch3:(1)The expressions of A and B in two groups were lower than those in group C at the end of 8th weeks(P<0.01,0.05).(2)The expression level of A group was lower than that of group B at the end of the 8th week(P>0.05).2.BMP-2:((1)The concentrations of A and B in the two groups at the end of the eighth week were higher than those in group C(P<0.01).(2)at the end of the 8th week,the concentration of A group was lower than that of group B(P>0.05).3.TGF-β:(1)The concentrations of A and B in the two groups at the end of the 5th week were higher than those in the C group(P<0.05,0.01).(2)At the end of 5th and the 8 week,the concentration of A group was lower than that of group B(P>0.05).Conclusion: 1.the occurrence and progression of SANFH was positively correlated with the expression of Notch3 protein,but wasnegatively correlated with the expression of BPM-2 and TGF-β.Huoxue Tongluo Decoction can down regulate Notch3 gene expression and the rate of empty bone lacunae femoral head,up regulating the expression of BMP-2 and TGF-β.It is suggested that Huoxue Tongluo Decoction can promote the differentiation and proliferation of osteoblasts and hematopoietic cells,inhibit the activity of osteoclasts and bone resorption.2.Huoxue Tongluo Decoction on prevention and treatment of SANFH may be related to inhibition of Notch3 gene and up regulate the expression of BMP-2 and TGF-β to induce BMSCs into osteoblasts,proliferation and differentiation and formation of cartilage cells and hematopoietic cells,and reduce the stimulation effect of hormones on the proliferation of broken bone cell differentiation then,inhibit bone resorption,delay bone necrosis of femoral head and promote local bone tissue repair and regeneration.3.This project is based on the "Chinese disease prevention first" characteristics of the guiding ideology,application of Huoxue Tongluo Decoction in SANFH before the intervention and Study on maintenance treatment for late model disease related experimental study and clinicaltreatment,provide new experimental treatment mode.
