Objective:Inthis study,the phosphorylation of progesterone receptor(PR)serine(Ser)190(Pho-PRser190),Forkhead box OlA(FOXOIA)and Forkhead box 03A(FOX03A)were detected by immunohistochemistry in 23 cases of Umbrellaof normal fallopian(UNF),23 cases of ovarian serous borderline tumor(OSBT),25 cases of Low grade ovarian serous carcinoma(LOSC)and 25 cases of High grade ovarian serous carcinoma(HOSC).To investigate the role of Pho-PRser190,FOXOIA and FOX03A in the pathogenesis and development of ovarian serous carcinoma and its relationship with clinicopathological features.Methods:The expression of Pho-PRser190,FOXOIA and FOX03A in UNF,OSBT,LOSC and HO SC were detected by Immunohistochemical Maxvision method.Image analysis software(Image-ProPlus)analysed the average mean optical density(MOD);SPSS18.0 statistically analysed Positive rate and the average brightness density value.Results:1.Comparison of the positive rate and intensity of Pho-PRser190,FOXOIA FOX03Ain UNF,OSBT,LOSC and HOSC.1.1 The positive rate of Pho-PRser190 in UNF,OSBT,LOSC and HOSC were 65%,96%,72%and 46%.The OSBT group was higher than the UNF group(p<0.05),and there were no statistically significant difference in the positive rate between LOSC and UNF,HOSC and UNF;Average MOD of Pho-PRser190 in UNF,OSBT,LOSC and HOSC were 0.2634,0.2244,0.1291 and 0.1155.Both the LOSC group and HOSC group were lower than the UNF group(p<0.05),but there was no statistically significant difference in average MOD between OSBT and UNF.1.2 The positive rate of FOXOIA in UNF,OSBT,LOSC and HOSC were 91%,78%,96%and 86%.There were no significant difference in positive rate between OSBT and UNF,LOSC and UNF,HOSC and UNF;Average MOD of FOXOIA in UNF,OSBT,LOSC and HOSC were 0.2153,0.1487,0.2394 and 0.1384.HOSC group was lower than UNF group(P<0.05),but there were no significant difference in average MOD between OSBT and UNF,LOSC and UNF.1.3 The positive rate of FOXO3A in UNF,OSBT,LOSC and HOSC were 91%,91%,92%and 89%.There were no significant difference in positive rate between OSBT and UNF,LOSC and UNF,HOSC and UNF;Average MOD of FOXOIA in UNF,OSBT,LOSC and HOSC were 0.3458,0.2771,0.1997and 0.1566.LOSC group and HOSC group were lower than UNF group(P<0.05),but There was no significant difference in MOD mean between OSBT and UNF.2.The results of Pearson correlation analysis MOD of Pho-PRser190,FOXOIA and FOXO3Ain LOSC and HOSC showed that there were no correlation between them.3.Relationship between average MOD of Pho-PRser190,FOXOIA,FOX03A and clinicopathological features of serous ovaran carcinoma.3.1 Relationship between expression average MOD of Pho-PRser190 and clinicpatho-logical features:Patients under 50 years of age was higher than over 50 years old(P<0.05);Patients with early clinical stage(?+? stage)was higher than those with advanced stage(?+? stage)(P<0.05);Patients without implant metastasis was higher than those with implant metastasis(P<0.05);The average MOD was not related to histological grade.3.2 Relationship between expression average MOD of FOXOIA and clinicopathological features:Patients under 50 years of age was higher than over 50 years old(P<0.05);Patients with early clinical stage(?+? stage)was higher than those with advanced stage(III+IV stage)(P<0.05);Patients without implant metastasis was higher than those with implant metastasis(P<0.05);The patients with LOSC was higher than those with HOSC(P<0.05).3.3 Relationship between expression average MOD of FOX03A and clinicopathological features:Patients under 50 years of age was higher than over 50 years old(P<0.05);The average MOD were not related to clinical stage,implant metastasis and histological grade.Conclusion:The decrease in Pho-PRser190 may cause the alteration of the target gene and participate in the occurrence of OSC.The decrease of expression intensity of FOXOIA may influence the expression of tumor suppressor gene,apoptosis of cells,signal pathway and so on,which involves in the occurrence of HO SC.The decrease of expression intensity of FOXOA may influence the expression of tumor suppressor gene,apoptosis of cells,signal pathway and so on,which involves in the occurrence of OSC.Pho-PRser190,FOXO1A,FOXO1A and FOX03A may be an important effective molecule in OSC.
