Objective:To establish and use gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid?Gd-EOB-DTPA?enhanced MRI to assess a novel rat model of heterogeneous hepatic injury.Methods:Seventy male Sprague-Dawley rats were randomly divided into control group?n=10?and colchicine group?n=60?.A dose of 0.4ml/kg with 0.25%colchicine was injected via the splenic vein to develop a rat model of heterogeneous hepatic injury.An equal volume of normal saline was injected via the splenic vein in the control group.After the operation,seven rats were randomly selected in the colchicine group at Day 3,week 1,week 2,week 4 and ten rats at week 8 and week 12 for MRI examinations after anesthesia prior to being euthanized.Rats in the control group underwent MRI examinations after anesthesia at Day 3,week 1,week 2,week 4,week 8,and week 12after the operation and were euthanized after MRI examinations at week 12.The detailed scanning settings were T2-weighted images?T2WI?,diffusion weighted imaging?DWI?,gadoxetate disodium enhanced MRI.MRI Image Analysis was performed to obtain LMR2from T2WI,?LMR from gadoxetate disodium enhanced MRI and ADC value from DWI both of the left and right liver lobes and liver histopathology was studied.Results:There was obvious pathological change of rat liver tissue after colchicine injection through splenic vein.The difference in hepatic injury induced by colchicine between the left and right lobes was significant at each time point?P<0.05?.Our data did reveal a statistical significance in ADC?×10-3 mm2/s?differences of the left and right liver lobes?Day 3:0.110±0.121 vs-0.001±0.005;week 1:0.091±0.027 vs 0.010±0.010;week 2:0.234±0.087 vs 0.009±0.011;week 4:0.171±0.053 vs 0.011±0.008;week 8:0.173±0.058 vs 0.035±0.022;week 12:0.190±0.078 vs 0.021±0.011;P<0.05?,LMR2 differences of the left and right liver lobes?Day 3:0.85±0.045 vs 0.016±0.011;week 1:0.067±0.015 vs 0.007±0.004;week 2:0.081±0.027 vs 0.008±0.006;week 4:0.110±0.062 vs 0.007±0.005;week 8:0.090±0.040 vs 0.006±0.007;week12:0.092±0.046 vs 0.007±0.005;P<0.05?,as well as?LMR differences of the left and right liver lobes?Day 3:0.182±0.054 vs 0.014±0.007;week 1:0.140±0.044 vs0.015±0.007:week 2:0.149±0.079 vs 0.016±0.009;week 4:0.234±0.095 vs0.015±0.007;week 8:0.171±0.061 vs 0.013±0.005;week 12:0.163±0.085 vs 0.012±0.004;P<0.05?between colchicine group and control group at each time point.It showed that LMR2,ADC value and?LMR decreased as the liver injury score increased and the correlations were statistically significant respectively?r=-0.245,P=0.018;r=-0.682,P=0.000;r=-0.807,P=0.000?.The correlation coefficient between?LMR and liver injury score was significantly higher than that between LMR2,ADC value and liver injury score.Conclusion:Colchicine injection through splenic vein can develop a successful rat model of heterogeneous hepatic injury.Gd-EOB-DTPA enhanced MRI is potentially useful for estimating heterogeneous liver injuty.
