| Objective: To explroe the different expression of leukemic inhibitory factor and its receptor and leptin in Decidua Tissue and serum progesterone level during early pregnancyMaterials and methods 1.In the obstetric ward and abortion outpatient department of the first affiliated hospital of China Medical University during April.2016-April.2017,30 cases of spontaneous abortion patients were treated as observation group 1,30 patierns had a history of taking progesterone and developed miscarriage as observation group2,30 cases of normal pregnancy as control group.Decidua and whole blood were collected during abortion.All patients signed the informed consent and passed the examination and approval of the ethics committee of the First Affiliated Hospital of China Medical University.2.The serum levels of progesterone were detected among the 3 group.3.TRIZOL method was used to extract the total RNA from 3 groups of decidua tissues.The Real-time PCR was used for exploring the expression of LIF,LIFR and 4.Western-blot was used to explore the different expression of LIF,LIFR and LeptinExperimental results: 1.the three groups had no statistical difference in gestational age,age,age,weight,height,BMI index,blood group,age of menarche,menstrual cycle,and the number of days.2.the serum progesterone level in the control group(81.31 + 7.52nmol/L)was significantly higher than that in the observation group 1(29.63 + 6.49 nmol/L)and the observation group 2(32.15 + 4.32 nmol/L),the difference was statistically significant(P < 0.05);there was no significant difference between the observation group 1 and the observation group 2 serum progesterone level.3.Western-blot and RT-PCR detection showed the relative expression of LIF(leukemia inhibitory factor)in the observation group 1 and observation group 2was significantly lower than the normal control group,with statistical difference(P < 0.05);the relative expression of LIF in observation group 1 and observation group 2 had no significant difference(P > 0.05).4.Western-blot and RT-PCR detection showed that the relative expression of LIFR in the observation group 1 and observation group 2 was significantly higher than the normal control group,with a statistically significant difference.(P < 0.05)5.Western-blot and RT-PCR detection showed that the expression of Leptin in the the observation group 1 and the observation group 2 was significantly lower than that of the normal control group(P < 0.05);the expression level of 1 in the observation group was significantly lower than that in the observation group 2(P< 0.05).Conclusion: The oral progesterone did not change the serum progesterone level and the protein and m RNA expression of LIF and LIFR in the decidua tissue,but up regulated the protein and m RNA levels of Leptin.It is suggested that abnormal pregnancy(low serum progesterone level)may not be improved by exogenous progesterone supplementation,resulting in a low expression of LIF... |
