Objective: bladder cancer with the highest occurrent rate and the most complicated pathogenesis,has been regarded as one of the lethal threaten to healthy of human in tumors of urinary system.It is illustrated from our previous results that ankyrin repeat protein(Diversin)which has been interested by our group play a significant role in tumorigenesis,malignancy,invasion and metastasis and that expression level of Diversin in carcinoma tissue of bladder cancer is significantly higher than it in paar-carcinoma tissue.What is more,our previous data shows that Diversin increase the capability of invasion and metastasis in non-small-cell Lung cancer via increasing the activity of Wnt/ JNK signaling pathway.Consequently,this project is aimed to confirm the function of Diversin in tissue level,to observe the efficiency of diversin in T24 cell line which induce the alteration of validity and capability of invasion and metastasis,and to explore the potential molecule mechanism of Diversin contributing cell proliferation.Method: Tumor samples were collected from 40 patients with urothelial carcinoma of bladder who had have operations between the year form 2009 to 2015 in Central Hospital Affiliated To Shenyang Medical College.All samples were analysis with immunohistonchemical staining to check the expression level of Diversin.To further confirm the efficiency of Diversin to tumorigensis,invasion and metastasis,T24 cell line were regarded as the objects which was temporarily transfected with Diversin special si RNA to downregulate Diversin expression level.Our project was designed that T24 cell line was checked with MTS to observe the capability of cell proliferation,examed capability of invasion and metastasis by Transwell assay,and analysis the activity of canonical Wnt/?-catenin and uncanonical Wnt/JNK signaling pathways through western-blot.Results: 1.result of immunohistonchemical staining Diversin in carcinoam tiissue of bladder tissue is significantly higher than it in para-carcinoma tissue.2.MTS results shows that downregulation of diversin could increase the capability of cell proliferation of T24 cell line.3.Trnswell assay results indicated that Diversin could increase the ability of invasion and metastasis of T24.4.Western-blot assay revealed results that Diversin unpregulates expression level fold of p-JNK/JNK which indicates that Wnt/JNK signaling pathway in T24 while activity canonical Wnt/?-catenin was not found.Conclusions: Overexpression of Diversin is correlated to bladder tumorgenisis,TNM stage,invsion and metastasis.Diversin increase the capablity of cell proliferation and invasion of T24.Diversin increase ability of proliferation,invasion and metastasis of the T24 cell via trigger Wnt/signaling pathway.
