Objective:To observe the effect of salidroside on the myocardium of rats under different altitude,to observe the protection of salidroside on rat cardiomyocytes under different altitude conditions,and to explore the mechanism of salidroside on cardiomyocyte apoptosis in Wistar rats.Methods:According to the principle of random group the 60 Wistar rats divided into the Lanzhou elevation group,5000 m elevation group,8000 m elevation group,Rats in each group were treated with normal saline or Salidroside for 3 days before entering the plateau environment simulationcabin,the Lanzhou elevation group in the laboratory normal altitude of 24 hours,the remaining two groups were entered into5000 m and 8000 m plateau environment simulationcabin for 24 hours.When get out the cabin,blood tests were performed in a timely manner,after executed immediately remove heart and lungs,then observed the changes HE staining in the rats cardiomyocytes and muscle fibers under immunohistochemistry.Using the Immunohistochemical method(IHC)to detect the expression of Caspase-3 gene in rat heart under different elevation conditions.Using the TUNEL to detect myocardial cell apoptosis.The expression of Bcl-2,Bax,Caspase-3 and Caspase-9 in rat cardiomyocytes were detected by Western Blot and WB.Results:1.The cardiac pathological staining results of rats in each group: There was no significant change in the myocardium of the rats in the saline group.The pathological changes were observed in the myocardium of the rats at the altitude of 5000 m and8000m respectively.The main manifestations were myocardial cell swelling,vacuolar degeneration and dissolution Necrosis and myocardial interstitial broadening,myocardial lesions under 8000 m altitude significantly greater than 5000 m altitude,the salidroside group rats myocardial cell morphology decreased compared with the saline group,myocardial cell enlargement and myocardial interstitial broadening degree Compared with the saline group improved.2.Myocardial apoptotic cells in each group: there were a few positive staining atLanzhou physiological saline group and a large number of positive staining cells at5000 m and 8000 m altitude.There were more positive cells at 8000 m than5000m,Under the altitude of Lanzhou,similar to that of the normal saline group at Lanzhou,the number of staining positive cells at 5000 m and 8000 m altitude was lower than that of the normal saline group and statistically significant compared with the saline group.(P <0.05)3.The results of the detection of Bcl-2,Bax,Caspase-3 and Caspase-9 in myocardial tissue of rats in each group :the expression of Bax,Caspase-3 and Caspase-2 expression decreased,Compared with the saline group,the expression of Bax,Caspase-3 and Caspase-9 in salidroside group decreased,while the expression of Bcl-2 increased,the difference was statistically significant.(P <0.05)Conclusion:1.High altitude acute myocardial hypoxia can lead to myocardial cell injury in rats.2.Salidroside injection can protect rat cardiomyocytes from injury under high altitude hypoxia,which may be related to the inhibition of the expression of Bcl-2,Bax,Caspase-3 and Caspase-9 in cardiomyocytes.
