Human Cytomegalovirus(HCMV)is one of the scale largest DNA herpescirus,its infection is widely spread in worldwide and cause serious human congenital disease.As an extremely dangerous virus,accurate diagnosis and effective prevention are very important.Currently,the plight of HCMV research is no effective diagnosis,no effective antiviral drugs and limited vaccines using.The development of animal models of HCMV infection was impeded by the strict species specificity of this virus.The existed animal models,such as mouse and Guinea pig,are established by using host's own cytomegalovirus.All these animal model couldn't represent the characteristics of human disease of HCMV infection.As a new experimental animal,tree Shrew has advantages of small size and homology similar to humans in many aspects,thus it is suitable to be taken as experimental animals.Herein,we expected to use tree shrew for establishment of cytomegalovirus infected animal model.Firstly,the primary lung cells was isolated from Tree Shrew,and cultured in vitro sucessfully.These obtained primary cells was inoculated using HCMV culture supernatants.It was showed that the cultured lung cell could be infected by HCMV and supported virus replication.Further,the lung cell was serious affected by virus infection presenting the cell deformation and death.Based on the obtained results from in vitro HCMV infection and previous reports,the infection dose of living Tree Shrew infected with HCMV was determined.Secondly,the clinical indicators were detected by nasal trip infection HCMV to Tree Shrew,body weight,blood routine and biochemistry were larger than normal healthy Tree Shrews,both urine and blood detected positive,including pulmonary focal hemorrhage,these were consistent of the clinical manifest actions of cytomegalovirus pneumonia.Finally,the lung was HCMV infection and latent organs by the tissue nucleic and protein detection,and mainly concentrated in alveolar lung cavity structure.In conclusion,the urine and blood are the standard method for detecting HCMV pneumonia.Nasal infection is a feasible experimental method for the cytomegalovirus pneumonia model of Tree Shrew.It can cause infection or lantent virus in the lung of Tree Shrew,these studies provide experimental and factual basis for the establishment of the model of HCMV infection by Tree Shrew.
