Objective:To study the expression of nucleolar protein(PES1,Pescadillo)in breast cancer tissues,analyze the relationship among PES1 andER,PR,HER-2,as well as discuss its significance in pathogenesis and prognosis of breast cancer.Methods:Surgical specimens of 60 patients with breast cancer(BC)were collected and fixed by 10% formalin and embedded.After routinely HE staining,typical tissue blocks were selected.Immunohistochemical staining was used to detect the expression of PES1 protein.The expression level of PES1 protein was analyzed with the patient's age,pathological type,histological grade,molecular classification,ER,PR,HER-2 and Ki67.Results:1.High expression of PES1 in breast cancer tissues:PES1 was mainly expressed in the nuclei of breast cancer tissues.In 60 cases of breast cancer,48 cases showed positive expression,and the positive expression rate was 80%(48/60).In 60 cases of paracancerous tissues,32 cases showed positive expression,and the positive expression rate was 53.3%(32/60).The expression of PES1 in breast cancer tissues was significantly higher than that in adjacent tissues,and the results were statistically significant(c2=9.6,P=0.020).The expression of PES1 in breast cancer tissues was significantly higher than that in adjacent tissues.2.The relationship between the expression of PES1 and clinicopathological features :1)The relationship between PES1 expression and age:In the age group between PES1 expression and agession was found in 25 cases(83.3%),PES1 negative expression in 5 cases(16.7%),in the age group >40 years old,PES1 positive expression in 23 cases(76.7%),and PES1 negative expression in 7 cases(23.3%)There was no correlation between the expression of PES1 in the two groups(c2=0.417,P=0.519).2)The relationship between the expression of PES1 and the pathological types of breast cancer:In breast infiltrating ductal carcinoma(IDC),PES1 positive expression was found in 25 cases(80.6%),PES1 negative expression in 6 cases(19.4%),in infiltrating lobular carcinoma(ILC),PES1 Positive expression was found in 15 cases(83.3%),PES1 negative expression in 3 cases(16.7%),in other types,PES1 positive expression in 8 cases(72.7%),PES1 negative expression in 3 cases(27.3%).In other cases,one case was solid papillary carcinoma of the breast and the other was mucinous adenocarcinoma of the breast.There was no correlation between the expression level of PES1 and histological type(P=0.390).3)Relationship between the expression of PES1 and the histological grade of breast cancer :In grade 1 of histological grade,PES1 positive expression was 12 cases(80.0%),PES1 negative expression was 3 cases(20.0%),in level 2,PES1 positive expression was 20 cases(80%),PES1 negative expression was 5 cases(20%).Grade 3,PES1 positive expression in 15 cases(83.3%),PES1 negative expression in 3 cases(16.7%),other refers to the incisive ductal carcinoma and invasive lobular carcinoma histological grading data inconsistent breast solid papillae In carcinomas and mucinous adenocarcinomas of the breast,PES1 positive expression was found in 1 case(50.0%)and PES1 negative expression in 1 case(50.0%).There was no correlation between the expression level of PES1 and histological grade(P=0.681).3.Correlation of PES1 expression with hormone receptor status,HER-2 status and Ki67 in breast cancer :1)Relationship between PES1 expression level and ER status :In the ER positive group,PES1 was positively expressed in 30 cases(90.9%),and PES1 was negatively expressed in 3 cases(9.1%).In the ER-negative group,PES1 was positively expressed in 18 cases(66.7%),and PES1 was negatively expressed in 9 cases(33.3%).There was a correlation between the expression of PES1 and ER(c2=5.455,P=0.020).2)Relationship between PES1 expression level and PR status :In the PR-positive group,28 cases(87.5%)were positive for PES1,and 4 cases(12.5%)were PES1 in the vagina.In the PR-negative group,PES1 was positively expressed in 20 cases(71.4%),and PES1 was negative in 8 cases(28.6%).There was no correlation between the expression of PES1 and PR(P=0.219).3)Relationship between PES1 expression level and HER-2 status :In the HER-2 positive group,PES1 was positively expressed in 30 cases(85.7%),and PES1 was negatively expressed in 5 cases(14.3%),in the HER-2 negative group,PES1 was positively expressed in 18(72.0%),and PES1 was negatively expressed in 7 cases(28.0%).There was no correlation between the expression of PES1 and HER-2(c2=1.714,P=0.190).4)Relationship between PES1 expression level and Ki67 :In Ki67 nship betw,25 cases(89.3%)were positively expressed in PES1 and 3 cases(10.7%)were negative in PES1.In Ki67<20% group,23 cases(71.9%)were positive for PES1 and 9 cases were negative for PES1(28.1%).There was no correlation between the expression of PES1 and Ki67(c2=2.829,P=0.093).4.The relationship between the expression of PES1 and the molecular subtypes of breast cancer :According to the St.Gallen Breast Cancer Consensus(2015)breast cancer is divided into at least four molecular subtypes.The positive expression of PES1 in Luminal A type was 10 cases(83.3%),PES1 negative expression was 2 cases(16.7%).Luminal type B PES1 was positively expressed in 14 cases(77.8%)and PES1 negatively expressed in 4 cases(22.2%).The positive expression of HER-2 overexpressing PES1 was 19 cases(86.4%),PES1 negative expression was 3 cases(13.6%).The positive expression of PES1 was found in 5 cases(62.5%)in three negative types,and negative in 3 cases(37.5%)in PES1.The expression level was not correlated with the molecular subtypes of breast cancer(P=0.543).Conclusion:The expression of PES1 protein in breast cancer tissues was significantly higher than that in adjacent tissues.There was a correlation between PES1 and ER expression,and there was no correlations with age,histological type,grade,molecular type,PR,HER-2,and Ki67 expression.The high expression of PES1 in breast cancer tissues and its positive correlation with ER expression suggest that PES1 may participate in the development of breast cancer through ER hormone pathway.
