PES1 Regulates Malignant Behavior Of Pancreatic Cancer Cells

Objective PES1 was overexpressed in PANC-1 and BxPC-3 pancreatic cancer cell lines to observe cell proliferation.The pancreatic cancer cell line stably expressing PES1 shRNA was constructed to study the effect of knocking down PES1 gene on the growth of pancreatic cancer cells,and further To investigate the effects of PES1 gene on invasion and migration of pancreatic cancer cells and tumorigenic ability in vivoMethods Using pcDNA3.1(+)as a vector,construct Flag-PES1 and transfect pancreatic cancer cell lines to explore the effect of overexpressing PES1 gene pancreatic cancer cells;Transfecting pancreatic cancer cells PANC-1 and BxPC-3 with virus packaging plasmid to obtain a virus that knocks down the PES1 gene,infecting the virus with pancreatic cancer cells and screening for stable clones of pancreatic cancer cells capable of stably expressing the knockdown PES1 gene.RT-qPCR was used to detect the expression of PES1 mRNA;the expression level of PEST protein was detected by Western blotting;the cell growth curve was used to detect the effect of knockdown of PES1 gene on the growth of pancreatic cancer cells;the transwell assay was used to detect the ability of knockdown of PES1 gene to pancreatic cancer cells.The effect of invasive ability;subcutaneous tumor formation in nude mice detected changes in the tumorigenic ability of pancreatic cancer cells after knocking down the PES1 gene.Results(1)overexpression of PES1 gene,the proliferation of pancreatic cancer cells was significantly higher in the experimental group than in the control group(P<0.05);(2)This experiment successfully obtained pancreatic cancer cells stably expressing the PES1 shRA gene;(3)Knockdown of PES1,the proliferation of pancreatic cancer cells were lower in the experimental group than in the control group(P<0.05);(4)Knockdown of PES1,the migration ability and invasive ability of pancreatic cancer cells were lower in the experimental group than in the corresponding control group(P<0.001);(5)Knockdown of PES1,the tumor volume and quality of the pancreatic cancer cells in the subcutaneous tumor formation of the experimental group were lower than the corresponding control group,and the Ki67 test group was lower than the corresponding control group(P<0.001).Conclusion PES1 gene can regulate the malignant behavior of pancreatic cancer cells,and the in-depth study of the regulation mechanism of PES1 in pancreatic cancer can find an effective way for molecular targeted therapy of pancreatic cancer.
