| Objective : To investigate the application value of insulin-like growth factor IImRNA binding protein 3(IMP3),glypican 3(GPC3)alone or combined detection in diagnosis of HCC.Methods:Detect the expression of IMP3 and GPC3 in 119 cases of HCC surgical specimens,50 cases of HCC biopsy specimens,4 cases of liver high grade dysplastic nodules(HGDN)specimens,6 cases of liver low grade dysplastic nodules(LGDN)specimens,10 cases of liver focal nodular hyperplasia(FNH)tissues and 10 cases of chronic hepatitis(CH)by using immunohistochemical method.Analyze the sensitivity and specificity of IMP3 and GPC3 alone or combined,and analyze the relationship between the expression of IMP3,GPC3 and clinical pathological features.Results : The positive expression of IMP3 in HCC surgical patients was 57.1%(68/119),the positive expression of HCC biopsy samples in the rate of 52%(26/50),the positive expression of GPC3 in HCC surgical patients was 83.1%(99/119),the positive expression of HCC biopsy samples in the rate of 56%(26/ 50).The sensitivity of IMP3 was 55.6%,the sensitivity of GPC3 was 75.1%,and the two combined sensitivity was85.7%,which is significantly higher than that of IMP3 and GPC3 individually.HCC specimens were further divided into well,moderatedly and poorly differentiated groups,it can be found that the difference of positive rate between joint detection and GPC3 is mainly in PDHCC.The expression of IMP3 has correlation with cirrhosis,pathological grade and serum AFP level(P = 0.027,0.012,0.005),while the expression of GPC3 was only related with serum AFP level(P=0.045).IMP3 and GPC3 were both not expressed in para-carcinoma tissues,HGDN,LGDN,FNH and CH.Conclusion : High expression of IMP3 and GPC3 were showed in the HCC organization.The sensitivity of joint detection was significantly higher than that of the single index,which is helpful for the diagnosis of HCC,so we recommend combine multiple indicators in the diagnosis of HCC to improve the accuracy. |
