Objective: Primary liver cancer is one of the most common malignancies in the world.The common histological type is hepatocellular carcinoma(HCC),which is a common cause of death due to cancer.Most of these cases and deaths are estimated to have occurred in China.Although important progress has been made in studies on its occurrence and development,the specific pathogenesis of HCC remains unclear.Myobuncin-1(Fascin1)is an actin binding protein found mainly in mesenchymal tissues and the nervous system.Recent studies have shown that Fascin1 is expressed in many malignant tumors and that its expression is significantly associated with tumor cell differentiation and metastasis,as well as patient prognosis.Phosphatidyl inositol proteoglycan-3(GPC3)can regulate the progression of malignant tumors through various pathways,and is closely related to tumor differentiation.Currently,GPC3 is widely used in the detection of early hepatocellular carcinoma,and it is a commonly used immunohistochemical index in clinical practice.This study focuses on the expression level of Fascin1 and GPC3 in HCC and their relationship with clinicopathological features and prognosis of HCC patients.Methods: This topic uses clinical research and database analysis combined method.(1)Clinicopathological data of HCC patients(including gender,age,HBsAg,cirrhosis,degree of tumor tissue differentiation,TNM stage,tumor diameter,capsule invasion,serum AFP concentration,hepatic microvascular invasion(MVI))were collected to obtain the survival status of patients.(2)The expression levels of Fascin1 and GPC3 in cancer and para-cancer tissues were tested by Immunohistochemical(IHC);(3)The difference of Fascin1 and GPC3 expression in cancer and para-cancer tissues by chi-square test;The relationship between the expression of Fascin1 and GPC3 and clinicopathological features by Chi-square test;The correlation of Fascin1 and GPC3 is analyzed by Sepearmen method.Spearman method was used to analyze the correlation of Fascin1 and GPC3 expression and clinical parameters.The prognosis was analyzed by Kaplan-Meier survival method.(4)Download the prognostic hotspot map of Fascin1 and GPC3 in different tumor tissues from GEPIA 2 database to analyze the impact of Fascin1 and GPC3 on the prognosis of HCC patients;Survival curve of Fascin1 and GPC3 for HCC patients was drawn,and Logrank P<0.05 was used to determine the difference in prognosis.Results:(1)The expression of Fascin1 was significantly higher in cancer tissue than paracancer tissue,with statistically significant difference(P<0.05).High expression of Fascin1 was associated with lower differentiation of HCC tissue and later TNM stage(P <0.05),but not with gender,age,HBsAg,tumor diameter,envelope invasion,serum AFP concentration,liver cirrhosis,and hepatic microvascular invasion(MVI)(P >0.05).(2)The expression of GPC3 in cancer tissues was significantly higher than that in paracancer tissues,and the difference was statistically significant(P<0.05).High expression of GPC3 was associated with lower differentiation degree of HCC tissues and later TNM stage(all P <0.05),but not with gender,age,HBsAg,tumor diameter,capsule invasion,serum AFP concentration,cirrhosis,and hepatic microvascular invasion(MVI)(all P >0.05).(3)Fascin1 and GPC3 expression in HCC tissue were positively correlated(R =0.483,P =0.001).(4)Kaplan-Meier survival analysis of clinical patients,compared with patients with high expression of Fascin1,the overall postoperative survival of patients with low expression of Fascin1 was longer,with statistically significant difference(Logrank P = 0.002).High or low expression of GPC3 was not associated with the prognosis of patients(Logrank P=0.321).According to GEPIA 2 database analysis,the overall survival of Fascin1 with high expression and low expression group has statistical significance(Logrank P=0.015),while the overall survival of GPC3 with high expression and low expression group has no statistical significance(Logrank P=0.2).Conclusion:(1)Fascin1 and GPC3 are up-regulated in HCC tumor tissue,which is related to the differentiation degree and TNM stage of tumor tissue.(2)Fascin1 expression in HCC tumor was positively correlated with GPC3 expression;(3)High expression of Fascin1 is a risk factor affecting prognosis of hepatocellular carcinoma patients.Therefore,it is speculated that upregulated Fascin1 and GPC3 can jointly promote HCC evolution,which may be related to its impact on biological behaviors such as tumor differentiation and invasion.Fascin1 may become a new indicator for co-detection of HCC with GPC3.Fascin-1 affects the prognosis of HCC patients,patients with high expression have a worse prognosis,and fascin-1 can be an important prognostic indicator of HCC patients.
