| Objective:To Explore the TLR4 gene 3′-non-coding regions rs11536889G/C polymorphism and the correlation of sepsis in children,for the early diagnosis and treatment of sepsis susceptible children provide genetic evidence and provide new ways of treatment.Methods: The sepsis group selected 96 cases of children who were admitted to our hospital from October2015 to October 2017 for the diagnosis of sepsis,including 51 males and 45 females,whose age rang from0.1~ 13.4 years,with an average age of(3.9±2.7)years old.The severity of sepsis was divided into 48 cases with sepsis,32 cases with severe sepsis and 16 cases of septic shock group.In the control group,80 cases of healthy children were selected,including 42 males and 38 females,whose age rang from 0.1 ~13.8years,with an average of 4.4 plus or 2.7 years of age,and collected clinical data.Case group and experimental group of children are all in the morning on an empty stomach take 2 ml peripheral venous blood,to-80℃ refrigerator.Set aside.Press kit instructions to extract DNA,in-20 ℃ low temperature,Genetic polymorphism of tlr4-rs11536889 gene was detected by design primer,PCR amplification target gene and gene level.Compare different severity of sepsis patients and healthy children TLR4 gene polymorphism have differences,analysis level of TLR4 gene polymorphism with sepsis in children and the relationship between disease severity,at the same time will be the basic clinical data were collected for statistical analysis.SPSS 17 software was used for data analysis,measurement data using the mean and standard deviation((?)± s),using t test or analysis of variance;chi square test with count data were compared;the correlation between variables by correlation analysis,2 test was used in calculating the allele frequency of the OR value and 95% confidence interval using Hardy-Weinberg,to verify the mass balance law of representative sample,the P>0.05 sample representative.Results:1.In the case group and the control group,there was no statistical difference between the two groups.2.Sepsis group,severe sepsis group and septic shock group between the three groups were compared: 22 age,sepsis and septic shock group age P<0.05;compared with the severe sepsis group and septic shock group age P<0.05;age differences were statistically significant,with age decreases.Sepsis is more and more serious;no significant gender differences between the three groups(P>0.05).3.The frequency of TLR4-11536889 locus conforms to Hardy-Weinberg equilibrium,insuring the reliability of their application to evaluate larger groups.4.There was a significant difference in TLR4-rs11536889G/C gene polymorphisms between the two groups(2==4.29 P=0.038).The risk of sepsis in TLR4-rs11536889 allele carriers was 1.474 times that of G allele carriers(95%CI:1.010 to 2.513).There was no significant difference in the polymorphisms of tlr4-rs11536889 in sepsis group,severe sepsis and septic shock group(P> 0.05).5.Conclusion:(1)The Gene polymorphism of TLR4-rs11536889 may be associated with the risk of sepsis in children.The C allele may be susceptible to sepsis,and carrying C may increase the risk of sepsis.(2)There was no significant correlation between gene polymorphism of TLR4-rs11536889 and the severity of children’s sepsis. |
