Objective: Discussing regulating hematopoiesis function and specific links of bone marrow stromal cells of preparate polygonatum decoction on hematopoietic system in the body,which is in “blood deficiency?consumptive disease” state caused by bone marrow suppression after radiation and chemotherapy;clearing the biological substance of polygonatum ' qualities of “ enriching deficiency and benefiting essence ” and the possible role of traditional Chinese herb of nourishing kidney essence in clinical treatment of tumor and blood disorder.Methods:The experimental research is based on myelosuppressive mouse models established by injecting Cyclophosphamide in abdominal cavity and infusing preparate polygonatum decoction into stomach.Using bone marrow stromal cells culture,MTT method,colony-forming unitfibroblast count,fluorescent real-time quantitation PCR,Western Blot,alkaline phosphatase staining and ELISA method and so on to observe the shape of bone marrow stromal cells,adherence time of80%,colony-forming unit-fibroblast numbers,the expression of hematopoiesis regulation factors GM-CSF and TNF-a,the expression of adhesion molecule ICAM-1 protein,the activity of alkaline phosphatase,the change of osteocalcin content and compared with normal saline group,discuss the effect and mechanism of Rhizoma Polygonati on bone marrow stromal cells in inhibition of hematopoiesis mice with multilevel and systematic.Results:Firstly,Preparate polygonatum decoction can promote the proliferation of bone marrow stromal cells,cut down the adherence time of 80% and promote the formation of colony-forming unit-fibroblast numbers of bone marrow stromal cells in inhibition of hematopoiesis mice caused by cyclophosphamide.Secondly,Preparate polygonatum decoction group can increase the content of ICAM-1 protein of bone marrow stromal cells when compared with normal saline and model group in inhibition of hematopoiesis mice caused by cyclophosphamide,.Tirdly,Preparate polygonatum decoction can advance the activity of alkaline phosphatase and the content of osteocalcin of bone marrow stromal cells when compared with normal saline and model group in inhibition of hematopoiesis mice caused by cyclophosphamide.Fourthly,Preparate polygonatum decoction can't change the expression of GM-CSF m RNA and TNF-a m RNA ofbone marrow stromal cells in inhibition of hematopoiesis mice caused by cyclophosphamide.Conclusion:Firstly,Preparate polygonatum decoction can promote the proliferation of bone marrow stromal cells in inhibition of hematopoiesis mice caused by cyclophosphamide,which can repair of damaged hematopoiesis induced microenvironment,and then reconstruct hematopoietic function.Secondly,Preparate polygonatum decoction can enhance the adhesion of bone marrow stromal cells in inhibition of hematopoiesis mice caused by cyclophosphamide,which can enhance the adhesion between bone marrow stromal cells and hematopoietic cells,and then reconstruct hematopoietic function.Tirdly,Preparate polygonatum decoction can increase related the indexes of the osteoblasts,which promoting bone marrow stromal cells in inhibition of hematopoiesis mice caused by cyclophosphamide to be osteoblast.It is the major component of HIM and can secrete many hematopoietic regulatory factors and adhesion molecules to regulate hematopoiesis.It may be one of the mechanisms of Preparate polygonatum decoction to regulate hematopoiesis.
