Objective: Discussing the effect and the underlying mechanism of preparate polygonatum decoction on hematopoietic system in the body,which is in “blood deficiency、consumptive disease” state caused by bone marrow suppression after radiation and chemotherapy;clearing the biological substance of polygonatum’ qualities of“enriching deficiency and benefiting essence” and the possible role of traditional Chinese herb of nourishing kidney essence in clinical treatment of tumor and blood disorder.Methods: The experimental research is based on myelosuppressive mouse models established by injecting Cyclophosphamide in abdominal cavity.Using blood cell count,organ weighting,MTT method,cell culture,hematopoietic progenitor colony assay,fluorescent real-time quantitation PCR,to observe preparate polygonatum decoction’ effect on peripheral blood cell,hematopoietic organs,proliferation of bone marrow cells,adherence ability of bone marrow stromal cells,progenitor colony forming rate,the m RNA expression of the kidney EPO and the spleen EPOR respectively.Results: The present research shows:1、Different doses of preparate polygonatum decoction by gavage in mice can increase the number of RBC、Hb、Plt and WBC in dose-dependent manner.The number of RBC、Hb、Plt began to rise from the 7th after doses and the WBC increase from the 14 th.The Hb content of NS group is significantly lower than that of model group each point in time,as the RBC in the 14 th.2、Each dose group of preparate polygonatum decoction could increase the index of thymus and spleen from the 14 th.And the index of spleen of middle and high dose group is significantly higher than that of low dose group.The organ index of high dose group is still higher than that of NS group on the 10 th and 14 th and higher than that of low dose group on the 14 th.The spleen index of middle group is higher than that of NS group on the 14 th.3、The proliferation of bone marrow cells of different doses of preparate polygonatum decoction is higher than that of NS group showing a trend of dose dependent on the 7th、10th and 14 th.From the 7th to14 th,the OD value of high dose group rise from 0.142±0.007 to 0.174±0.005,and that of NS group from 0.121±0.007 to 0.126±0.004.4、The adhesion ability of BMSC of high dose group is 27.55% on the10 th,and that of model group is 19.45%.5、The rate of CFU-E、BFU-E、CFU-GM、CFU-Meg of myelosuppressive mice improve significantly after lavaged with high dose of preparate polygonatum decoction,showing a remarkable difference with NS group.6、The secretion of Epo in kidney of high group in 10 th is nearly three times as much in NS group,and the expression of EpoR in spleen rose showing a significant different with NS group.7、All the detection indexes of NS group are lower than that of model group each point in time,especially the Hb of each point 、RBC in 14 th and CFU-E in 10 th decreased significantly.Conclusion: The present research approves:1、Preparate polygonatum decoction could improve the proliferation of hematopoietic progenitor cells by improving the proliferation of bone marrow cells,prompting the proliferation and differentiation of hematopoietic stem cells to different types hematopoietic progenitor cells.Thus it contributes to the recovery of peripheral blood in myelosuppressive mice caused by injecting Cy.But the onset time on different cells may be different.It increase the number of RBC through promoting the secretion of Epo in kidney and improving the expression of EpoR in spleen.2、Preparate polygonatum decoction can improve the adhesion ability of bone marrow stromal cells,further enhance the function of hematopoiesis induced microenvironment.Finally it promote the recovery of hematopoietic.3、Preparate polygonatum decoction increase the index of thymus and spleen,so it has effects on the immunity functionmye in suppressive mice,4、Long-term gastric perfusion maybe cause damage to experimental animals body.So setting the lavage group with normal saline in experiment can hedge the system error.
