Study Of The Effects On Fear Memory Of PTSD Rats Intervened By Calming Shen-Restoring Consciousness-Regulating Kidney Electro-acupuncture Therapy Based On BDNF-TrkB-ERK Pathway In Amygdala

Objective:1.To observe and compare the behavioral effects of SPS method and SPS&S method on rats with PTSD.2.To observe the effects of electroacupuncture on the fear memory acqusition,extinction and reconstruction of PTSD rats3.To observe the effects of electroacupuncture on the the BDNF-TrkB-ERK signal pathway in amygdala of PTSD rats for reveal the central mechanism of electroacupuncture therapy for PTSD.Methods:After adaptive breeding,50 male SD rats was randomly divided into 5 groups:control group;SPS model group;SPS with electroacupuncture group;SPS&S model group;SPS&S with electroacupuncture group.Each group had 10 rats.We used the method of SPS and SPS&S to make symptoms for the rats except the control group.EA treatment(on the points including BaiHui,ShenTing,Shen Shu)for the EA groups.After the treatment,we use conditioned fear reaction test to detect the rats behavioral change.The expression of BDNF,TrkB,MEK,ERK protein,number of immunoreactive cells and mRNA in amygdala were detected by using IHC and Western-blotting respectively.SPSS23.0 and GraphPad Prism 7 software are used for data analysis and cartography.Results:1.Obeservation and comparison of the effectiveness of SPS and SPS&S methods in modeling PTSD rats1)Scene conditioned fear response detection results show that compared with Ctrl group,there was a significant difference in the scene conditioned fear response of SPS&S group and SPS group in the acuqsition,extinction and reconstruction stage.Compared with SPS group,there was a significant difference in the scene conditioned fear response of SPS&S group.2)Cue conditioned fear response detection results show that compared with Ctrl group,there was a significant difference in the cue conditioned fear response of SPS&S group and SPS group in the acuqsition,extinction and reconstruction stage.Compared with SPS group,there was a significant difference in the cue conditioned fear response of SPS&S group.2.Effect of electroacupuncture on fear memory acuqsition,extinction and reconstruction of PTSD rats1)Scene conditioned fear response detection results show that there was a significant difference in the scene conditioned fear response of SPS&S group and SPS&S+EA group in the extinction and reconstruction stage.2)Cue conditioned fear response detection results show that there was a significant difference in the cue conditioned fear response of SPS&S group and SPS&S+EA group in the extinction and reconstruction stage.The results showed that the electroacupuncture could significantly improve the extinction and reconstruction of fear memory in PTSD rats.3.IHC resultsCompared with Ctrl group,BDNF(p<0.01),TrkB(p<0.01),p-ERK(p<0.01)of SPS&S group decreased significantly(p<0.01),while protein expression level of BDNF(p<0.01),TrkB(p<0.01),p-ERK(p<0.01)in SPS&S+EA group was significantly up-regulated(p<0.01).4.Western Blot resultsCompared with Ctrl group,BDNF(p<0.01),TrkB(p<0.01),p-MEK(p<0.01),p-ERK(p<0.01)of SPS&S group were significantly reduced,while SPS+EA group can significantly increase BDNF(p<0.01),TrkB(P<0.01),p-MEK(p<0.01),p-ERK(p<0.01)level of protein expression.Conclusion:1.Both PTSD rat models made by SPS and SPS&S meet the requirements of fear memory sensitivity animal model.2.Electroacupuncture can improve the clinical core symptoms of PTSD rats and improve the fear memory extinction and reconstruction.3.Electroacupuncture can upregulate the expression of BDNF,TrkB,p-MEK,p-ERK in the amygdala of PTSD rats.4.Electroacupuncture significantly improve the acquisition,regression and reconstruction of fear memory in PTSD rats by activating the BDNF-TrkB-ERK signaling pathway.
