Objective:To detect the role and mechanism of 1,25-dihydroxyvitamin D3 in renal tissue of rats with unilateral ureteral obstruction.To observe the pathological changes of rat kidney and to detect the expression levels of alpha-smooth muscle actin,interleukin-17and Phosphorylation alpha-AMP-activated protein kinase protein in renal tissue.Methods:Healthy male Wistar rats were selected and randomly divided into 3 groups:sham operation group,UUO group and UUO+1,25?OH?2D3 intervention group,each group of 18.In the UUO group and the intervention group,right ureter ligation was performed.The intervention group was given gavage of 1,25?OH?2D3 3ng/100g/d,and the other two groups were given the same amount of peanut oil.The rats were sacri ficed at days3,7 and 14 for extraction of the right kidney.Observed the renal pathological changes by HE staining.The levels of IL-17,?-SMA and p-AMPK?protein were detected by Western blot.Results:Compared with the sham operation group,the protein expression of?-SMA and IL-17 in UUO group increased significantly and the protein expression of p-AMPK?decreased significantly?all P<0.05?.Compared with the UUO group,the protein expression of?-SMA and IL-17 in the intervention group was significantly decreased and the protein expression of p-AMPK?was significantly increased?both P<0.05?.Conclusion:1,25?OH?2D3 delayed the progression of renal interstitial fibrosis,and this effect may be related to inhibit inflammatory through activation of the AMPK?pathway.
