| Objective:To observe Toutongning Capsule to nitroglycerin cause migraine in rats serum dopamine(DA),dopamine beta hydroxylase(D?-H),norepinephrine(NA)content influence,and to investigate the prevention and cure effect of Toutongning Capsule on migraine,and the possible mechanism.Methods:1.36 healthy male Wistar rats normal were fed for a week,randomly divided into blank control group(6 rats)and migraine model group(30rats),and blank control group given saline subcutaneous injection,migraine model group by subcutaneous injection of nitroglycerin(NTG)preparation of migraine model rats,and randomly divided into model control group,prevention of high dose group,prevention of low dose group,high dose treatment group,low dose treatment group.Blank control group: The distilled water was given by intragastric administration2ml/kg/d for 7 days,and seventh days after gavage for 30 minutes,10ml/kg saline was injected subcutaneously.Model control group: The distilled water was given by intragastric administration2ml/kg/d for 7 days,and seventh days after gavage for 30 minutes,10ml/kg NTG was injected subcutaneously again.Treatment of high/low dose group: The distilled water was given by intragastric administration 2ml/kg/d for 7 days,and seventh days after gavage for 30 minutes,10ml/kgNTG was injected subcutaneously again,after 30 minutes of the model,760mg/kg and380mg/kg Toutongning suspensions were separately given by intragastric administration.Prevention of high/low dose group,Toutongning suspensious was separately given by intragastric administration 2ml/kg/d for 7 days,and seventh days after gavage for 30 minutes,10ml/kg NTG was injected subcutaneously again.2.Then observe the change of behavior of rats and record the number of times when rats scratch head,bite toe head / tail,round-trip action and climb cage in every time period.3.Serum samples from rats were collected at 30 minutes,60 minutes and 180 minutes after the completion of the experimental procedure.The contents of DA,D?-H and NE were measured by enzyme linked immunosorbent assay(ELISA).Results:1.Behavioral changes: the behavioral changes of rats were observed after the last administration in different periods of symptom(scratch head,bite toe head / tail,round-trip action and climb cage etc.),blank control group after the last given saline,climb cage and bite toe head / tail action number within 30 minutes is more than the times of 30 to 60 minutes and 150-180 minutes,may be associated with subcutaneous injection of saline irritability.Migraine related behaviors in prevention of high/low dose group were significantly lower than those in model control group(P < 0.05);treatment of high dose group were lower than those in model control group(P < 0.05),and there was no significant difference in the related behavior of migraine in the low dose group compared with the model control group(P > 0.05).2.DA,D?-H,NE content changes: through the single factor analysis of variance,6groups DA,D?-H,NE content change was statistically significant(P < 0.05),The multiple comparison of the data of each group showed that prevention of high dose group(60 and180 minutes)of serum D?-H content compared with the normal control group had no statistical significance(P > 0.05),prevention of high dose group(60 minutes)of serum D?-H content compared with the normal control group had no statistical significance(P >0.05),comparion between two any means in other groups were statistically significant(P< 0.05).3.The correlation of DA,D?-H and NE content: there is a negative correlation between DA and NE,and there is a negative correlation between DA and D?-H,and NE has a positive correlation with D?-H.Conclusion:Toutongning capsule may through increase the activity of D?-H,so as to reduce the content of DA,increase NE content,give play to the role of preventive and treatment of migraine,and its effect on migraine prevention than treatment. |
