Experimental Anxiety Model In Rat Brain Tissue. The Bushen Ningxin Particles Norepinephrine, Dopamine Affect Experimental Research

Purpose: Using the conflict principle that the instinct fear and the inquisition actuation of animals under the novel environment, we use the elevated plus-maze test( EPM) to invigorate the anti-anxious function of Bushen Ningxin pellets from the behavior study, explore the treatment mechanisms of Cardiac neurosis by observing the effect of norepinephrine (NE), dopamine (DA) in experimental anxiety model rats'brains .Method:1 .60 male Wistar rats, weighing 180±10g, were randomly divided into blank group, DZP Comparison group, model group, Bushen Ningxin pellets high dose group, Bushen Ningxin pellets middle dose group and low Bushen Ningxin pellets dose group, 10 rats in each group.2. Bushen Ningxin pellets high-dose group, middle dose group, low-dose group were gavaged daily from different concentrations of Bushen Ningxin suspension particles, DZP Comparison group were gavaged diazepam tablets 1mg/kg/d, the model group and blank group were gavaged equal volume of normal saline. Gavage for 7 days.3 .After the 7th day gavaged to finished, in addition to the blank group, other groups made the behavior study test.After the end of study, rats were killed rapidly decapitated to remove the hypothalamus setting on ice .4 .By enzyme linked immunosorbent assay(ELISA) to detect content of the NE, DA in the hypothalamus of each group rats, observed the effect of the Bushen Ningxin pellets.Result: Model group, NE, DA concentration was significantly higher than blank group (P <0.01); Compared with model group, DZP Comparison group and Bushen Ningxin pellets high-dose group of NE, DA concentrations were significantly different (NE: DZP Comparison group P <0.001, Bushen Ningxin pellets high-dose group P <0.01; DA: DZP Comparison group P <0.01, Bushen Ningxin pellets high-dose group P <0.01); Bushen Ningxin pellets high-dose group Compared with the blank group and DZP Comparison group ,there was no significant difference (P> 0.05).Conclusion:Bushen Ningxin pellets can reduce the content of NE, DA in the hypothalamic,by reducing or inhibiting NE, DA release to reduce their impact on the cardiovascular system, played the role of the treatment of Cardiac neurosis.
