Objective:To observe the expression effect of adenomyosis mice the effects of estrogen related factors PTEN and P-P65 by endometriosis rehabilitation has the efficacy of Qingshi Huayu,Immunohistochemical technique was used to detect the expression of the above factors in the uterus of mice in each group before and after treatment,To explore the mechanism of clearing away dampness and removing blood stasis in the treatment of adenomyosis.Method: The 8 week old ICR rats were selected,using the method of pituitary transplantation.Then the mice were randomly divided into high,middle and low dose groups of NYKFP,DEFKJG,traditional Chinese medicine control group,GES,western medicine control group,March model group,June model group,Sham operation group,Blank control group.There were 24 mice in each group,Conventional feeding 3 months,The mice in the march model group,the blank group and the sham operation group were sacrificed during estrus and estrous cycle on average.June model group continued to feed.The remaining mice who were given corresponding drugs,were sacrificed during estrus and estrous cycle on average 3 months later.The expression of PTEN and P-P65 in the uterus of mice was detected by immunohistochemistry.The pathological changes of adenomyosis before and after the treatment were observed by HE staining and pathological sections,The severity of AM was graded.Result :1.From the morphological observation,the histological grading of adenomyosis and the severity of adenomyosis were observed,There was no significant difference between sham operation group and normal group(P>0.05),The March model group and the June model group were significantly higher than those in the normal group in the year of March,Statistically significant(P<0.01),June model group was higher than the March model group,No statistical significance(P>0.05)。2.There was no significant difference in the expression of PTEN and P-P65 between normal group and sham operation group(P>0.05),The expression of P-P65 increased gradually with the progression of AM in the model group,June model group and the March model group were significantly higher than those in the normal group and sham operation group(P<0.01),June model group was higher than the March model group.The expression of PTEN decreased gradually,June model group and the March model group were significantly lower than those in the normal group and the sham operation group(P<0.01),June model group was lower than the March model group(P<0.01)。3.The expression of P-P65 was decreased and the expression of PTEN was increased after treatment in AM mice.High dose groups of NYKFP,DEFKJG group,GES group,June model group were statistically significant(P<0.01),The expression of P-P65 of high dose NYKFP group was significantly lower than that of GES group and GES group(P<0.01),The expression of PTEN of high dose NYKFP group was significantly higher than that of GES group and GES group(P<0.01),Different doses of endometriosis rehabilitation between any two groups were statistically significant.4.Whether it’s estrus or estrous cycle,The expression of estrogen related factor P-P65 in ectopic endometrium of AM model mice was higher than that in eutopic endometrium(P<0.01),Eutopic endometrium was higher than normal endometrium,The difference was statistically significant(P<0.01),The expression of PTEN was lower than that in eutopic endometrium(P<0.01),Eutopic endometrium was lower than normal endometrium,The difference was statistically significant(P<0.01),And the expression of PTEN and P-P65 in the normal group and the model group of mice with different stages of the estrous cycle changes,The expression of P-P65 in estrus was significantly higher than that in estrous(P<0.01),The expression of PTEN in estrus was significantly lower than that in estrous(P<0.01).Conclusion:1.The operation process had no effect on the modeling results,The disease severity score increased with the duration of disease.In this study,the success rate of the AM mice model was established after the transplantation of the same type of pituitary gland after 3 and 6 months,and the success rate was up to 100%,This method is worthy of popularization and Application.2.The occurrence of adenomyosis is related to the abnormal expression of PTEN and P-P65 in the local estrogen related factors,This study suggests that P-P65 expression is up-regulated,and the down-regulation of PTEN expression may lead to the occurrence of adenomyosis.3.The traditional Chinese medicine of clearing away dampness and removing blood stasis can improve the expression of PTEN and play a role in the treatment of adenomyosis by reducing the expression of estrogen and estrogen related factors P-P65.However,the mechanism of regulating PTEN and P-P65 needs further experimental study.4.The expression of high dose NYKFP and DEFKJG and GES can reduce AM eutopic and ectopic endometrium effect related factors of P-P65,and increase the expression of PTEN.High dose NYKFP is more effective than two kinds of drug control group,NYKFP’s ability to regulate PTEN and P-P65 is related to does,The effect of high dose group was higher than that of middle dose group and the effect of middle dose group was higher than that of low dose group.The experiment shows that the treatment of adenomyosis by the method of removing dampness and removing blood stasis is worthy of further study and popularization.5.In this experiment,the expression in estrus of P-P65 was significantly higher than that in estrous cycle(P<0.01),and the expression in estrus of PTEN was significantly lower than that in estrous cycle(P<0.01).The method of clearing away dampness and removing blood stasis has no obvious effect on the change of periodicity.This will provide the basis for choosing the best time for treatment of adenomyosis.
