The famous-region drug is the index of evaluate and control quality of Chinese crude drug.The research of two Sichuan authentic ingredients,Penthorum chinense Pursh.and Lonicera similis Hemsl.have been discussed in view of our research group.The amount of novel lignins were isolated from the ethyl acetate part of Penthorum chinense Pursh.and hepatoprotective activity was screened by chemical constituents and systemic pharmacological experiments.Comparison analysis on TLC,HPLC,GC-MS and FT-IR fingerprints of different strains of Lonicera similis was conducted by multiple analytical means available.The research contents of this paper include two objectives:On the one hand,to investigate the chemical constituents and establish the quality standard from Chinese traditional medicine Penthorum chinense Pursh.On the other hand,to investigate the chemical constituents and their antitumor activities from Chinese traditional medicine Lonicera sirmilis Hemsl.The main results are as follows:(1)The compositions(P1-P12)of Penthorurm chinense Pursh.were isolated and purified by various chromatographic methods including column chromatography over silica,Sephadex LH-20,and Rp Ci8 columns.Their structures were identified by physicochemical properties and spectroscopic analysis.The compound numbers,names and structures have shown in table 1-1 and figure 1.1.On the basis of earlier researches of chemical constituents and pharmacological activities,ten batches of Penthorum chinense Pursh.in Sichuan and Guizhou provinces were collected to establish the TLC with Pinocembrin-7-O-?-D-glycosidase and Penthorin A as comparison samples,establish the methods for determinations of content with Pinocembrin-7-O-?-D-glycosidase,Thonningiania B,PGHG,Thonningiania A as comparison samples,establish the fingerprint of Penthorum chinense Pursh.and identify eleven peaks,and stipulate the moisture,ash,acid-insoluble ash,extractum standards of Penthorumchinense Pursh.The draft was draw up a document based on the above researches to provide a reference for the quality control.(2)The compositions(Ll*-L23)of Lonicera similis Hemsl.were isolated and purified by various chromatographic methods,including five lupane-triterpenoid saponins(Ll*-L5),eight hederagenin-triterpenoid saponins(L6-L12),two bisepoxylignans(L13?L14),five molecular phenolic acid compounds(L17-L22).Their structures were identified by physicochemical properties and spectroscopic analysis.The compound numbers,names and structures have shown in table 1-1 and figure 1.1.The anti-proliferative effect of HEPG-2,MCF-7,A-549 were determined by MTT,in which IC50of L12 were 8.98±0.19?M?12.48±0.45 ?M?11.62±0.54 ?M respectively.The results showed that L12 perfomed significant antitumor effect,and other compounds were no active.Furthermore,Hoechst fluorescence 33342 staining were used to demonstrate that 12 could induce HEPG-2 cells and A-549 cells.Based on the current data,it can conclude that 12 induces cell toxicity in HEPG-2 and A-549,in which apoptosis plays a very important role.
