Objective:Observing the different therapeutic effects of the single acupoint and double acupoints in the treatment of insomnia model rats on Shenmen and Sanyinjiao,and exploring the effect of electroacupuncture at different acupoints on LC-NE system and PG-MT system in rats with insomnia.Methods:Divideding the healthy and clean grade SD rats into 5 groups:blank group,model group,Shenmen group,Sanyinjiao group,Shenmen+ Sanyinjiao group.Model group,Shenmen group,Sanyinjiao group and Shenmen +Sanyinjiao group were established model successively for 15 days to duplicate the model of insomnia animal by restricting activity method,except as the blank group which was fed normally.Each group had the 24 h locomotion activity test,the elevated plus-maze test and the rat tail suspension test after the modeling.After the examination,the Shenmen group and Sanyinjiao group rats were treated on single point of Shenmen and Sanyinjiao,and the Shenmen + Sanyinjiao group rats with the treatment of Shenmen and Sanyinjiao double acupoints,successively for 5 days,but the model group was synchronizing fixed without treatment.Each group had the 24 h locomotion activity test,the elevated plus-maze test and the rat tail suspension test after the treatment,enzyme linked immunosorbent assay was used to detect the content of NE and MT in the plasma of rats in each group,and the positive expression of NE and MT in brain tissue was detected by immunohistochemistry.Results:(1)Behavior after modeling:(1)The 24 h locomotor activity:Compared with the blank group,The daytime activity of model group,Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group increased significantly(P<0.05),and the night activity reduced significantly(P<0.05);(2)The elevated plus-maze test:The model group,Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group which compared with blank group are significantly reduced in the OE%,OT% and the total distance movement,central entry,central time(P < 0.05).(3)There is nosignificant difference between the five groups in the static time of tail suspension experiment(P>0.05).(2)Behavior of different combinations of Shenmen and Sanyinjiao after treatment:(1)The 24 h locomotor activity:The daytime activity of model group is significantly higher than the blank group(P < 0.05),and the night activity is significantly decreased(P<0.05);After electroacupuncture treatment,the daytime activity of Shenmen group,Sanyinjiao group and Shenmen + Sanyinjiao group compared to model group is significantly decreased(P<0.05),and the night activities are significantly increased(P<0.05),but there is no significant difference compared with the blank group(P > 0.05);There are no significant difference between the Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group(P>0.05).(2)The elevated plus-maze test:the model group is significantly lower than the blank group in the OE%,OT% and the total distance movement,central entry,central time(P<0.05).After electroacupuncture treatment,the Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group compared to model group is increased significantly(P<0.05),but there is no significant difference compared with the blank group(P>0.05);There are no significant difference between the Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group(P>0.05).(3)There is no significant difference between the five groups in the static time of tail suspension experiment(P>0.05).(3)The content of NE and MT in the plasma of different combinations of Shenmen and Sanyinjiao after treatment:(1)NE content in the plasma:The model group was significantly increased compared with the blank group(P<0.05);After electroacupuncture treatment,the Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group compared to model group is decreased significantly(P<0.05),but there is no significant difference compared with the blank group(P>0.05);There are no significant difference between the three treatment groups(P>0.05).(2)MT content in the plasma:The model group is significantly less than the blank group(P<0.05);After electroacupuncture treatment,the Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group compared to model group is increased significantly(P<0.05),but there is no significant difference compared with the blank group(P>0.05);There are no significant difference between the three treatment groups(P>0.05).(4)The positive expression of NET in the LC and MT1 in the PG of different combinations of Shenmen and Sanyinjiao after treatment:(1)Positive expression of NET in the LC:MOD of the model group is significantly higher than the blank group(P<0.05);After electroacupuncture treatment,MOD of Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group is significantly lower than model group rats(P<0.05),but there is no significant difference compared with the blank group(P>0.05);There are no significant difference between the three treatment groups(P>0.05).(2)Positive expression of MT in the PG:The MOD of the model group is significantly less than the blank group(P<0.05);After electroacupuncture treatment,the MOD of Shenmen group,Sanyinjiao group and Shenmen+Sanyinjiao group compared to model group is significantly increased(P<0.05),but there is no significant difference compared with the blank group(P>0.05);There are no significant difference between the three treatment groups(P>0.05).Conclusions:The single or double acupoints of Shenmen and Sanyinjiao with electroacupuncture all can improve the disorder sleep and alleviate anxiety of insomnia model rats,And the effect of treatment of insomnia in the single acupoint and double acupoints of Shenmen and Sanyinjiao show that there is no significant difference.Electroacupuncture on acupoints of Shenmen and Sanyinjiao is mainly by reducing the NET expression in the LC and peripheral plasma NE content of insomnia rat,while the positive expression of MT1 in the PG and peripheral plasma MT content is raised at the same time,then adjusting the function of sleep activation and inhibition system so as to achieve the therapeutic effect of insomnia.
