Study On The Effects Of Compatibility Of Shenmen And Sanyinjiao Regulating Central 5-HT1AR And GABA Of Insomnia Model Rats

Objective: Observing the influence of single point and combination of Shenmen(HT7)and Sanyinjiao(SP6)on the sleep-wakefulness,emotional state,and central 5-HT1 AR and GABA of insomnia model rats,to definite the effects of compatibility of Shenmen(HT7)and Sanyinjiao(SP6)treating insomnia and to explore its central mechanism,providing experimental research evidence for acupuncture treatment of insomnia.Methods: Selecting qualified experimental rats from healthy grade SD rats according to data of the body weight,open field test,elevated plus-maze test and rat tail suspension test,and dividing them to control group,model group,Shenmen group,Sanyinjiao group and ShenmenSanyinjiao group.The control group was fed normally,and the other groups were modeled by restricting activity.Each group had the 24 h locomotion activity test,open field test,and rat tail suspension test after the modeling.Then,the treatment groups were treated with electroacupuncture for 5 consecutive days.The model group was captured simultaneously without any treatment.After the treatment,each group had the 24 h locomotion activity test,open field test,and rat tail suspension test.Finally,the expressions of 5-HT1 AR in raphe nuclei,5-HT1 AR and GABA in suprachiasmatic nucleus were detected for each rat by immunohistochemistry technique.And the contents of 5-HT and GABA in plasma were detected by ELISA test.Results:(1)Five groups of rats in the body weight,peripheral lattice score,central square score and vertical score in the open field test,and stationary time in the rat tail suspension test were no significant difference before the modeling(P>0.05).(2)After the modeling,five groups of rats in the daytime activity,the nighttime activity,the peripheral lattice score,central square score and vertical score in the open field test had statistic difference(P<0.05),in which the model group and treatment groups compared with the control group,the rats’ daytime activity,central square score and vertical score had obviously increased,decreased in the nighttime activity and peripheral lattice score(P<0.05).The stationary time in the rat tail suspension test had no significant difference between each group(P>0.05).(3)After the treatment,five groups of rats in the daytime activity,the nighttime activity,the peripheral lattice score,central square score and vertical score in the open field test had statistic difference(P<0.05).The model group compared with the control group,the rats’ daytime activity,central square score and vertical score had obviously increased,decreased in the nighttime activity and peripheral lattice score(P<0.05).The treatment groups’ daytime activity,central square score and vertical score had significantly decreased,increased in the nighttime activity and peripheral lattice score,compared with the model group(P<0.05).The stationary time in the rat tail suspension test had no significant difference between each group(P>0.05).(4)After the treatment,five groups of rats in the contents of 5-HT and GABA in plasma had statistic difference(P<0.05).The model group compared with the control group,the rats’ content of 5-HT in plasma had obviously increased,decreased in the content of GABA in plasma(P<0.05).The treatment groups’ content of 5-HT in plasma had significantly decreased,increased in the content of GABA in plasma,compared with the model group(P<0.05).(5)After the treatment,five groups of rats in the mean density of 5-HT1 AR in raphe nuclei,5-HT1 AR and GABA in suprachiasmatic nucleus had statistic difference(P<0.05).The model group compared with the control group,the rats’ mean density of 5-HT1 AR in raphe nuclei,5-HT1 AR and GABA in suprachiasmatic nucleus had obviously decreased(P<0.05).The treatment groups’ mean density of 5-HT1 AR in raphe nuclei,5-HT1 AR and GABA in suprachiasmatic nucleus had significantly increased,compared with the model group(P<0.05).Conclusions:(1)Insomnia model rats are characterized by sleep disorder,and anxiety,which can be evidently improved with electroacupuncture at each or both of Shenmen(HT7)and Sanyinjiao(SP6).(2)The expressions of 5-HT1 AR and GABA in central nervous system and the content of GABA in plasma have obviously decreased,increased in the content of 5-HT in plasma,of the insomnia model rats with dysfunction of ascending reticular activating system and ascending reticular inhibiting system.Electroacupuncture can improve this and make the insomnia model rats recover to normal sleep cycle.(3)The effects of single point and combination of Shenmen(HT7)and Sanyinjiao(SP6)on improving insomnia model rats’ sleep and emotional state have no evidently difference.
