The subject focused on healthy Sprague Dawley(SD)male rats,which were randomly divided into equally ten groups,respectively normal control group(saline),model group(saline),positive drug control group(silymarin),7 given drug groups(different ratios of 18?-Gly and 18?-Gly group,and ratios are 10:0,8:2,6:4,5:5,4:6,2:8,0:10)once daily for four weeks before administration of 40% alcohol.The objective was to investigate the effect of different proportions of 18?-and 18?-glycyrrhizic acid(Gly)on liver functions,the histology of organs and inflammatory cytokines in rats with alcoholic liver injury.Firstly,Alcoholic liver disease(ALD)was discussed,including clinical studies survey and the pathogenesis,to clarify the purpose and significance of the study.Secondly,the objective of this part was that investigating protective effects of different proportion of 18?-and 18?-Gly on alcoholic liver function in rats with ALD.After the fourth week,blood was collected to determine serum ferment and proteins,total bilirubin and acid bile.Blood glucose was measured.The result illustrated that different proportions of 18?-and 18?-glycyrrhizic acid all effectively reduced ALT,AST,?-GT and LDH activity.The ratio was less than 4:6 that can better improve alcoholic liver injury and myocardial injury.The ratio was greater than 8:2 that improved serum protein level in alcoholic liver injury rat.The radio was less 4:6 that attenuated the level of TBIL and TBA.And the proportion was less 4:6 that obviously improved the ability of regulating the blood glucose of rats with alcoholic liver injury.It can be applied to selection of new drug research and clinical treatment with a basic experiment data.Thirdly,the objective of this part was to investigate the effect of different proportions of 18?-and 18?-Gly on the histology of important organs in rats with alcoholic liver injury.After fourth week,important organs were collected,including liver,kidney and spleen.Then organs were weighted and calculated organ indexes,and were stained by routine oil red O and hematoxylin-eosin.The histology of organs was observed by optical microscope.The results showed that 18?-Gly and 18?-Gly at the ratios of 2:8 significantly decreased organs indexes of respiratory and digestive system,obviously reduced muscle index and increased brain index,and meanwhile decreased cardiac index and spleen index.Additionally,18?-Gly and 18?-Gly at the ratios of 4:6 and 2:8 reduced hepatocytes steatosis and lipid accumulation.18?-Gly and 18?-Gly at the radio of 4:6 or less could decrease inflammation of the kidney,and meanwhile,alleviated injury to spleen demonstrated by the clear boundary of red and white pulp of spleen.Collectively,the optimal compatibility proportion of 18?-Gly and 18?-Gly on protecting organ injury induced by alcohol was 4:6 and 2:8.Fourthly,the objective of this part was to investigate the effect of different proportions of 18?-and 18?-Gly on the inflammatory cytokines and chemokines in rats with alcoholic liver injury.After fourth week,blood was collected from the abdominal aorta to test levels of cytokines and chemokines by Bio-Plex Pro TM.The results showed that 18?-Gly and 18?-Gly at the ratios of 4:6 and 2:8 significantly decreased levels of TNF-?,IL-1? and IL-6.Additionally,18?-Gly and 18?-Gly at the ratios of 5:5 or less reduced levels of IL-4,IL-5 and IL-7.18?-Gly and 18?-Gly at the radio of 4:6 could decrease chemokines levels comprehensively.All ratios of 18?-Gly and 18?-Gly could not significantly change the level of IL-10.18?-Gly was more likely to increase RANTES level.To sum up,the optimal compatibility proportion of 18?-Gly and 18?-Gly on decreasing alcohol-induced inflammation by alcohol was 4:6.The objective of this paper was to investigate the effect of different proportions of 18?-and 18?-glycyrrhizic acid(Gly)on different aspects in rats with alcoholic liver injury.And it provided experimental data support for preparation of clinical medication and would open the door to novel agents in new drug discovery.
