This article explored the effects of different proportions of glycyrrhizic acid epimer on the liver protection and lipid metabolism on ALD rats,further explored the molecular regulatory mechanism of the different proportions of glycyrrhizic acid epimer on ALD rats at lipid metabolism,and selected the best proportion of glycyrrhizic acid epimer.Chapter 1: This chapter introduced the progress of alcoholic liver injury and epidemiology,and lipid metabolism of alcoholic liver injury,analyzed the feasibility of glycyrrhizic acid treatment of ALD,and clarified the purpose and significance of the study.Chapter 2: This chapter investigated the effects of 18? and 18?-glycyrrhizic acid on the glucose metabolism,lipids metabolism and proteins metabolism in ALD rats.18? and 18?-glycyrrhiza acid improved glucose metabolism,lipid metabolism,and protein metabolism in ALD rats.And 18?-Gly is more effective in regulating glucose metabolism,lipid metabolism,and protein metabolism.The results will provide experimental basis for guiding rational drug use in clinics and developing new drugs.Chapter 3: This chapter studied the effect of different proportions of glycyrrhizic acid epimer on lipid metabolism in ALD rats.The serum,liver and feces TC,TG,HDL,and LDL levels were measured.The serum and liver SOD,GSH,and MDA levels were measured.And liver tissue was stained with HE and oil red O.The results showed that glycyrrhizic acid epimers significantly reduced serum and liver lipid levels,increased fecal lipid levels,reduced lipid accumulation,and significantly increased serum and liver SOD and GSH levels reduce MDA levels.Different glycyrrhizic acid epimer proportions have different regulatory effects on lipid metabolism in ALD rats.The optimal proportion is 4:6 on lipid metabolism in ALD rats.Chapter 4: This chapter studied the mechanism of different proportions of glycyrrhizic acid epimer on lipid metabolism in ALD rats.The mRNA and protein expression levels of SREBP-1c,ACC-1,PPAR-? and CPT-1 were determined by RT-PCR and Western-blot.The results showed that different proportions of glycyrrhizic acid epimers significantly inhibited the mRNA and protein expression of SREBP-1c and ACC-1,and significantly promoted the expression of PPAR-? and CPT-1 mRNA and protein.The optimal proportions of 18?:18?-Gly were 8:2,4:6,2:8 and 18?-Gly.Therefore,glycyrrhizic acid epimers can protect lipid metabolism disorders by inhibiting lipid synthesis and promoting lipid oxidation.
