Objective:To study the effect of Sinomenine Hydrochloride on the lipopolysaccharideinduced serum inflammatory factors and the degree of corneal endothelium injury in rabbits,in order to supply some experimental evidence to clinical practice on preventing one from getting arteriosclerosis and the treatment of systemicinflammatory response syndrome even to the major cardiovascular diseases.Methods:In this experiment,1-2 kilos of healthy New Zealand rabbits(female and male rabbits each half)to be randomly divided into six groups,including:control group,model group,SIN-L group,SIN-M group,SIN-H group and DEX group,5 in each group.All the rabbits received continually injection of drugs or normal saline with microperfusion pump.Combine LPS with stock solution at a dose of 0.5 milligrams per kilogram to build an experimental model for inflammation and endothelial injury in rabbits.The Sinomenine bind with its stock solution to be intervened at a dose of 20,40 and 80 milligrams per kilogram respectively in rabbits.Meanwhile,the Dexamethasone at a dose of 0.5 milligrams per kilogram to be intervened.Adopt the automatic biochemical analyzer for routine blood test,the quantification of blood circulating endothelial cells by the Percoll density gradient separation;Meanwhile,adopt ELISA to mensurate TNF-? and v WF's content in serum,finally,HE staining were taken to observe the morphological changes of cardiac and hepatic tissue.Results:According to the studies,the results show that:(1)Compared with control group,the count of peripheral blood leucocyte were obviously reduced in the model group(P<0.05);Contrast to model group,the SIN-M group's and the DEX group's peripheral blood leucocyte were recovered considerably(P<0.05),there had no differences between the SIN-L and SIN-H group in peripheral blood leucocyte despite both of them were increased a few(P>0.05);Contrast to control group,there had no differences of the recovery in peripheral blood leucocyte on SIN-M group(P > 0.05),while the DEX group were increased considerably(P<0.05),besides compared with SIN-M and DEX group,it is shown that the two has obvious change in peripheral blood leucocyte(P < 0.05).(2)Compared with control group,the count of circulating endothelial cells on rabbits were increased considerably in the model group(P<0.05);the results of three group of SIN and model group is compared,it turns out that the SIN-M group's circulating endothelial cells number obviously reduced(P<0.05),the other two groups also have a different degree of reduction in circulating endothelial cells,but has not significant(P>0.05);Contrast to model group,the count of circulating endothelial cells in DEX group also reduced significantly(P<0.05);but Compared with control group,there had no differences between the SIN-M and DEX group in circulating endothelial cells number aspect(P>0.05);it is shown that has no difference when compared two groups of their own(P>0.05).(3)Compared with control group,the level of serum concentration of inflammatory factor TNF-? and v WF were increased considerably in the model group(P<0.05);in a similar manner,Compared with model group,among the three group of SIN,the SIN-M group's level of serum concentration of inflammatory factor were obviously reduced(P <0.05),while the other two groups has the opposite results(P>0.05)and the DEX group also had obvious change(P<0.05).Compared with the group of SIN-M and DEX,there has no change in the level of TNF-?(P > 0.05)while the other two factors had obvious difference(P<0.05).(4)Compared with control group,the model group appeared hepatocyte swollen,denaturation,congestion of the liver and focal hemorrhage in the myocardium;Compared with model group,the SIN-M group's and the group of DEX could postpone the morphological change of heart and liver tissue while there has no significant changes between the other two groups;there were no differences in the changes of histomorphology between the SIN-M and DEX group.Conclusion:According to the experiment,we can draw the conclusion:(1)The Sinomenine could improve peripheral blood cell,which contain the leucocyte and the Circulating endothelial cells,induced by lipopolysaccharide and the effect of middle dose(SIN-M)was remarkable,at the same time,the effect could compare with dexamethasone to some extent.(2)The Sinomenine could reduce the level of serum concentration of inflammatory factor(TNF-? and v WF),then of the efficacy in a certain anti vascular endothelial injury so as to protect the vascular intima.(3)The Sinomenine could attenuate histopathological change in heart and liver,so we surmised that the Sinomenine may inhibit inflammatory factor,protect the body from inflammatory lesions.
