Objective:At present,the incidence of Chronic Fatigue Syndrome?CFS?is increasing at home and abroad.Especially in middle-aged people,CFS will lead to a serious decline in their quality of life.Therefore,research on prevention and control of CFS products has a good market prospect.Based on this,the study was based on the Jianzhi Fang?Polygalae Radix,Rehmanniae Radix Praeparata,Poria,Radix Asparagi?,aimed to preparing a tablets for improving the function of learning and memory reduction caused by CFS as a health care function,and their preparation process,quality standards and efficacy were studied.In this experiment,Polygalae Radix,Rehmanniae Radix Praeparata,Radix Asparagi and Poria were combined to treat the treatment of chronic fatigue syndrome in rats with reduced ability to learn and memory,and its therapeutic effect was initially explored.It is of great significance for the prevention and treatment of CFS and for the improvement of the quality of life of the CFS population.Methods:The factors including ethanol concentration,solid-liquid ratio,extraction time and extraction times were selected.The total saponins and total polysaccharide content of the active ingredients were used as indicators.The single factor experiment and L9?34?orthogonal design test were used to examine the extraction process of Jianzhi tablets.Then,the process of concentration and drying of the extract was investigated.After screening the type and amount of tablet molding excipients,the molding process of Jianzhi tablets was studied;the quality standards for Jianzhi tablets were established,including inspection,identification,and content determination.From swimming to physical fatigue and deprivation of sleep to mental fatigue,a rat model of chronic fatigue syndrome was established.After the administration,the rat Morris water maze experiment was performed,and the PITC pre-column derivatization HPLC technique was proposed to detect the contents of amino acid neurotransmitters?Glu,GABA?in each brain tissue?hippocampus,striatum,hypothalamus?.Results:The optimal extraction process of Polygalae Radix and Poria was as follows:7.5 times 60%ethanol was used to extract 3 times,and each extraction was 1.0 h;The optimal extraction process of Rehmanniae Radix Praeparata and Radix Asparagi was:using 10 times water to extract 3 times,and extracting 1.0hour each time.The concentrations of the alcohol extract concentrate and the water extract concentrate were 0.47g·mL-1and 0.32g·mL-1?raw drug weight:concentrated liquid volume?respectively.Concentrate was vacuum dried at 70°C to constant weight at-0.08-0.09Mpa.The optimal extraction process was as follows:the extract powders was sieved through 80 mesh and mixed.Add 25%total lactose,mannitol?2:1?and 2%CMS-Na,mix and spray 95%ethanol as wetting agent,granulation,dry at 60°C,and sieve over 16 mesh.Mesh whole,add 0.5%magnesium stearate,then mixed,pressed into piece.In the study of the quality standards of Jianzhi tablets,the results were all in compliance with the regulations,including the appearance of the appearance,usage and dosage,examination?weight difference,disintegration time limit?,identification and content determination,etc.In animal experiment,the results of the Morris water maze test showed that each group of Jianzhi Tablets could significantly improve the rats'navigation ability and space exploration ability caused by the CFS model.The results of determination of neurotransmitters?Glu and GABA?in brain tissue showed that compared with the model group,each dose group of Jianzhi tablets could significantly reduce glutamate content and increase GABA content,making it tend to normal level.Conclusion:The preparation process of Jianzhi Tablets is stable and feasible,and its quality standard is stable and reliable.It is a reliable experimental basis for industrialized production.Jianzhi Tablets can improve the learning and memory ability of CFS model rats by regulating Glu and GABA levels in the hippocampus,striatum and hypothalamus to normal levels,and maintaining a relatively balanced and stable Glu/GABA ratio.So as to improve the learning and memory ability.
