Gynura formosana Kitam.,the common name for Ganyancao,cool,sweet light,non-toxic,mainly into the liver,lung,kidney,large intestine.Mainly has anti-inflammatory,antipyretic,detoxification efficacy and so on.Widely used in folk of Fujian province especially Minnan in this area,generally used as a treatment of inflammation,especially hepatitis,has an admirable effect.It is a kind of valuable natural medicinal plants and herbal medicine resources.There is no reported about Gynura formosana Kitam.due to the use of it has remained among the folk.To clear and definite Gynura formosana Kitam.pharmacognostical characteristics,create a comprehensive and effective quality evaluation system,ensuring the safety and effectiveness of drug and expand the search for a new pharmaceutical resource provided a scientific basis.In this study,Gynura formosana Kitam.were studied systematically.Objeetive:Conducting a system of pharmacognosy identification of the Gynura formosana Kitam.,which will provide a scientific basis for its identification.At the same time,the extraction technology of total flavonoids,which is the main chemical composition in Gynura formosana Kitam.being optimized.Evaluating the protective effect of extracts from Gynura formosana Kitam.on acute liver injury and nonalcoholic fatty liver disease.Methods:By the method of pharmacognosy study the plant morphology,medicinal characteristics,microscopic characteristics(organization structure,powder characteristics of different medicinal parts)of Gynura formosana Kitam..Determined the concentration of ethanol,ethanol dosage,extraction time,extraction times in four factors level range by using the single factor experimental results.Measured the total flavonoids as indicators,used orthogonal experimental method to optimize the extraction parameters of its main chemical components.To observe the protective effects of Gynura formosana Kitam.on rat liver damage induced by carbon CC14,this study also established the rat model of nonalcoholic fatty liver disease to observe the role of Gynura formosana Kitam.in nonalcoholic fatty liver disease.Result:Well identificated rousuihong is compositae by the original plant roots,stems,leaves and powder trait identification,microscopic identification,which has the characteristics of stems cylindrical,rough hairy,leaves shallow crack,heads tidbits with fragrance,without glandular hairs,infinitive porosity,cork layer with stone cell band,powder has secretion and inulin.Optimized the extraction technology of total flavonoids as its main chemical components that is ethanol concentration 60%,30 times dosage of ethanol,extracting 3 times,each time 1 h.Pharmacodynamics research shows that Gynura formosana Kitam.alcohol extract has marked protective effects on acute liver damage induced by CCl4 in rats,has obvious anti effect of nonalcoholic fatty liver disease,can prevent excessive liver fat deposition to a certain extent,improve liver function and liver steatosis,alleviate,liver tissue inflammation.There is significant inhibition on the formation of fatty liver caused by high-fat diet in experimental rats.ConclusionThis study determined the Gynura formosana Kitam.base source,properties,microscopic characteristics,can provide reference for Gynura formosana Kitam.of crude drug identification.Extraction process optimized results for further technological development has a favorable reference value.Efficacy of pharmaceutical active study show that Gynura formosana Kitam.has significant inhibition on the formation of fatty liver caused by high-fat diet in experimental rats.And provided reference for its reasonable exploitation and utilization.
