Objective:To investigate propentofylline(PPF)impact on the plantar incision model of acute pain and spinal glial inflammatory factors.Methods:96 adult male SD rats weighing 250-300g,were randomly divided into 4 groups(n = 24),group ?(blank control group):Anesthesia,intrathecal catheter and injection of 0.9%N.S10ul;group?(surgical control group):0.9%N.S10ul after intrathecal injection of anesthesia plantar incision surgery pain model;group ?(preoperative intrathecal injection of PPF group):After intrathecal catheter,intrathecal injection PPF10ug/lOul first,then plantar incision surgery pain model;groupIV(after intrathecal injection of PPF group):After intrathecal catheter,plantar incision surgery pain model first,and then injected dose intrathecal PPF.1d ago were established in incisional pain model,after the establishment of 2h,6h,24h,48h,96h were observed mechanical pain threshold and heat pain threshold of the rats,after the behavioral experiments,take L4-6 spinal cord.Determination of spinal microglial marker OX-42,and the expression level of astrocyte marker GFAP by Western blot.Determination the expression of IL-1?,TNF-a and substance P by immunohistochemistry,and statistical analysis.Results:?,?,IV group compared with I group at all time points show hyperalgesia,mechanical pain threshold and thermal pain threshold are lower(P<0.05);in the rat spinal cord OX-42,GFAP,IL-1?,TNF-? and substance P expression are increased(P<0.05);?,IV group compared with group II hyperalgesia are abate,mechanical pain threshold and thermal pain threshold are higher;in the rat spinal cord OX-42,GFAP,IL-1?,TNF-? and substance P expression are reduced(P<0.05).? group compared with ? group,2 h,6 h,24 h,48 h after surgery,hyperalgesia are abate,mechanical pain threshold and thermal pain threshold are higher(P<0.05);in the rat spinal cord OX-42,GFAP,IL-1?,TNF-a?and substance P expression are reduced(P<0.05).Conclusion:Propentofylline can effectively alleviate the rat acute hyperalgesia toe incision and have analgesic action in advance.Its mechanism is connected with drug inhibition of spinal cord glial cells OX-42 in advance and the expression of GFAP protein,and spinal cord of rats inflammatory factor IL-1?,TNF-? associated with a reduced the release of substance P.
