Coronary artery heart disease(CHD)is a common multiple cardiovascular disease in clinical,with the first incidence in the primary heart disease.Due to a high morbidity and mortality,serious threat to human life and health.With the international development of Chinese medicine,traditional Chinese medicine used in the prevention and treatment of cardiovascular diseases caused extensive concern in the pharmaceutical industry.Therefore,to choose curative medicine,in-depth study on prevention and control mechanism,a clear role in its efficacy and characteristics,for research and development drugs for treating cardiovascular disease has important significance and clinical value.Clinopodium chinense is perennial herb of Labiatae in Clinopodium Linn,its active ingredient contains a variety of chemical constituents,such as flavonoids,triterpenoids and glycosides,steroids,organic acids and aromatic,etc,with the pharmacological effects of bleeding and blood circulation,anti-inflammatory,dimmune,hypoglycemic,anti-bacterial,anti-tumor,anti-radia tion,and so on.Flavonoids have significant antioxidant activity,but much research is currently to prove it has antioxidant activity on the natural products of flavonoids,there are also some studies involve analyzing the relationship between the structure and oxidation resistance,but the few studies have reported the ways and mechanism in antioxidant action.While flavonoids was widespread concerned due to its effects in protect the cardiovascular,but so far,the mechanisms of protect cardiovascular are still not fully elucidated.Therefore,for the study of natural flavonoids,molecular mechanisms of myocardial injury which were based on an important path to against oxidative stress has important scientific significance for revealing significant antioxidant intrinsic molecular mechanisms.To take a in-depth study that based on the levels of molecular and cellular to research the CTF protective effect and its mechanism on coronary artery heart disease,in vivo and in vitro.To confirm antioxidant protection by cell viability,antioxidant enzyme activity and intracellular reactive oxygen species(ROS),to confirm antioxidant mechanisms by JC-1 and AnexinV/PI staining,Caspase3 fluorescence active potential and Western blot assay with detecting mitochondrial membrane,apoptosis and various proteins expression through H/R-induced H9C2 model,in vitro.Acute myocardial injury model induced by isoproterenol hydrochloride(ISO)in vivo,detecting of tissue enzymes(LDH/CK/AST)vitality,antioxidant enzyme activity and histopathological changes to confirm the antioxidant effects;myocardial tissue by immunohistochemistry,to detect the relationship between the antioxidant effect and paths by transmission electron microscopy and Western blot,in order to clear the mechanism in total flavonoids.Vivo and in vitro experiments support each other,to articulate protective effect and its mechanism of Clinopodium chinense total flavonoids on coronary artery heart disease,fully.The results show that Clinopodium chinense total flavonoids have the exact protective effect on coronary artery heart disease,the mechanism are closely related to oxidative stress,apoptosis and PI3K/AKT and MAPK signaling pathways.
