Currently,there are many available commercial product of Tripterygium wilfordii such as Tripterygium preparations Tripterygium glycosides tablet,triptolide tablets,triptolide total patch,which clinically used for the treatment of nephrotic syndrome,rheumatoid arthritis,systemic lupus erythematosus and other autoimmune diseases.However,there are the high incidences of adverse reaction and there is a big difference at adverece action and effect among different manufacturers.Making it safer and more effective in clinical is hot and difficulty in Tripterygium research.Because of the complexity of compositions and structures in Tripterygium,only triptolide,Wilforlide or other single-component composition are analyzed,which can not solve the problem fundamentally.This paper study is aim to look for the low toxicity and high effect goup by the spectrum efficiencyso as to provide data preparation for quality evaluation and process optimization.In the early suty,Tripterygium preparations fingerprints of seven manufacturers are established by HPLC.The fingerprint including 18 peaks is generated,which provide date for the later spectrum relationshipestablishment.The sudy of anti-inflammatory,immunosuppressive and toxicodynamics pharmacodynamic was carried out and the xylene-induced ear edema in mice and carrageenan-induced paw edema in rats models were adapted to evaluate the anti-inflammatory efficacy with the detection of swelling degree;By using Freund's complete adjuvant-induced immune suppression adjuvant arthritis pharmacodynamic model,the immunosuppressive agents Pharmacodynamic of Tripterygium extract was evaluated with tumor necrosis factor-?,interleukin-6 and interleukin-1 content of indicators.Liver and kidney toxicity studies were done with indicators of aspartate aminotransferase(ALS),alanine aminotransferase(ALT)and serum urea nitrogen(BUN).It was found that anti-inflammatory and immunosuppressive activity was different by using different indicators to evaluate the Tripterygium preparations extracts from different manufacturers.The result of evaluation indicated that there was difference in impact to liver and kidney function among seven manufacturers.The analysical result of fingerprints and efficacy indicates that the function strength has something to do with the number of chemical composition and content in extracts of tripterygium Tripterygium preparations.Finally,the spectrum-effect relationships between HPLC fingerprint and anti-Inflammatory activities were established by using the partial least squares multiple linear regression analysis.The analysis results showed that the contribution of 1?4?8?13?14?16 and 18 peaks in 18 peaks was larger tothe anti-inflammatory efficacy;the contribution of 1?2?3?4?5?6?7?9?10?11?12?14?17 and 18 peaks was larger to immunosuppression;the contribution of 1?2?5?8?9?10?12?13?14 and 18 peaks was larger to hepatotoxicity;the contribution of 1?2?6?10?11 and 18 was larger to renal toxicity.According to the results,we find the No.3?4?7?15?16?17 peaks make great contribution to the pesticide effect,the No.11?12?14 peaks have great pesticide effect and little toxicity,and the No.1?2?5?6?8?9?10?13?18 peaks are important to the pesticide effect as well as the toxicity.
