Tripterygium wilfordii is one of traditional Chinese medicine and have variousactivities, inculding anti-inflammatory, anti-tumor, immunosuppressive and anti-HIVactivities, etc.. Its pharmaceutical preparations are used for the clinical treatment ofrheumatoid arthritics, other inflammatory and autoimmune diseases. But due to itsside effects, there is a lot of limitation in clinical application. The group of TianjinMedical University established the production process of supercritical carbon dioxideextraction from T. Wilfordii. It was based on the analysis of bioactive components andremoved toxic compound. A reasonable and scientific method will be set up by theway of chromatographic, pharmacological and toxicological analysis through thisstudy of the quality control.The method of content determination was established by chromatographicexperiments, including the content of tripterine, triptolide and the total alkaloids. Wehave established the chromatographic fingerprint of supercritical carbon dioxideextraction of T. Wilfordii. 27 peaks were identified as evaluative peaks, 21 peaks ofthem were identified as active compounds with the Rt time of reference compoundsand HPLC-MS. The method is steady and accurate with a good repeatability and canbe used as a quality control for the SFE extract of T. Wilfordii.Further pharmacological and toxic experiment indicating that the best extractiontechnologcial conditions is viable, and it can achieve aim to reduce the toxicity andenhance the effectivity of T.wilfordii. The above supercritical extract has threefold tofourfold activity and about 50% toxicity comparison with market product of T.wilfordii.The quality-control of SFE extract of T. wilfordii is based on content control oftripotolide & tripterine, combining the relative variable of 27 peaks in its finger print chromatography.
