Objective Iodine excessive disorders is a series of thyroid dysfunction disease caused by excessive iodine intake.Many patients will show some nerve mental symptoms associated with functional change of brain nerve center.Monoamine neurotransmitter is kinds of neurotransmitter,containing some special structure which can directly affect human body.At present,the reasoning conclusion about the relationship between monoamine neurotransmitter and high iodine exposure is mainly based on two aspects:one is that changes of monoamine neurotransmitter can interfere with the self-regulation of hypothalamus-pituitary-thyroid axis,the other one is that secretion of monoamine neurotransmitter is sensitive to changes of iodine in body.Researches about iodine and monoamine neurotransmitter are more focused in the 20th century,iodine deficiency part.Researches about relation between high iodine and monoamine neurotransmitter for now are less.We will use high performance liquid chromatography with electrochemical detector via high iodine animal experiment and population survey to determinate concentrations of monoamine neurotransmitter like E,NE,DA,5-HT and 5-HIAA,discuss relation between high iodine and monoamine neurotransmitterMethods 1.According to prestudy,we find that the suitable HClO concentration of pretreatment on brain is 0.1mol/L,1mL per 100mg.The suitable HClO of serum is 5%(V/V),100?L HClO in 100?L serum.After pretreatment brain tissue and serum turned in refrigerated centrifuge(4?)at 10000r/min 15minutes.The optimal chromatographic conditions are as follows:column is ODS-3 C18.Mobile phase are phosphate buffer[45mmol/L NaH2PO4,35mmol/L citric acid,0.25mmol/L EDTANa2,2mmol/L NaCl,2.2mmol/L sodium heptanesulfonatem,using NaOH to adjust pH=4.0]:acetonitrile=95:5,flow rate is 1.0 mL/min,sample volume is 20?L,column temperature is 35?,voltage is+0.8V.2.In animal experiment of high iodine 25 rats were randomly divided into five groups.After six months of iodine intervention,collect artery blood before death and separate brain tissue,detect the contents of NE,E,DA,5-HT and 5-HIAAwith HPLC-ECD after perchloric acid processing.3.The population investigation is involved of high water iodine regions and optimal water iodine area.Compared with of normal adults,we will scan patients with hypothyroidism as a group.After collecting serum,we will detect NE,E,DA.5-HT and 5-HIAA with HPLC-ECD.Results 1.In animal experiments,NE in cerebral cortex,striatum and serum were decreased with the increase of iodine intake(P<0.05);E along with the increase of iodine intake in serum and decreased(P<0.05);the content of DA in striatum are presenting a peak type as iodine intake increased.Compared with 30.75?g group and 61.50?g group,DA of 307.50?g group and 615.00?g group decreased(P<0.05).5-HT of cortex in 61.50?g group are higher than control group and 30.75?g group(P<0.05),in 307.50?g,615.00 ?g group 5-HT are lower than 61.50?g group(P<0.05).In serum,5-HT in 307.50 and 615.00?g group are lower than 30.75?g and 61.50?g group(P<0.05);5-HIAA in cortex,striatum and serum present similar peak type change.2.In population survey,in high iodine water area,5-HT and 5-HIAA of hypothyroidism patients are higher than that of normal adults(P<0.05).In normal iodine water regions,NE of hypothyroidism patients less than normal adults(P<0.05),DA are not detected,5-HT and 5-HIAA are higher than normal adults(P<0.05).Conclusions 1.In animal experiments,NE decreased with iodine increased.NE is a kind of excited medicine,which can contract blood vessels and increase blood pressure.As the NE content reduces,the effect of NE also drops;it could be one of the reasons that IED patients may have some cardiovascular system damage.Disorders of DA can affect emotional reactions;it may be one of the reasons for abnormal emotional responses of IED patients.5-HT is associated with a variety of physiological functions.Disorders of 5-HT and 5-HIAA may explain IED patients may have some nervous system diseases,such as memory loss,depression,etc.2.In population survey,the hypothyroidism patients of two areas have higher 5-HT and 5-HIAA content than normal adults.It prompts that 5-HT system in hypothyroidism condition is metabolically active.All data above prompt ue that a series of mental illness and the balance appeared on hypothyroidism patients may be associated with monoamine neurotransmitter.Therefore,for hypothyroidism patients,we should focus on thyroid hormone as well as on monoamine neurotransmitter levels.We should try to treat thyroid disease and improve iodine nutritional status,at the same time,we also can permit intervention treatment according to different mental nerve symptoms and neurotransmitter content changes in serum on patients.
