| As a perennial evergreen herb and one species of Saxifragaceae plant,Bergenia crassifolia(L.)Fritsch is mainly produced in Siberia,Altai Mountains as well as central and eastern part of Asia.It mainly grows in Russia,Mongolia and Xinjiang Altay Mountains at an altitude of 300-2600 meters,which grows in cracks of rocks and forest.Bergenia crassifolia is a traditional medicine in Russia,Mongolia and Xinjiang.It is one of the common medicine in China.Its whole plant has pharmacological activities like antitussive,astringency and hemostasis.Kazak also used it to treat diarrhea,dysentery,tuberculosis,cough and other symptoms.In addition,Bergenia crassifolia medication in Xinjiang region has a long history,widely used in folk recipe.However,the research about the chemical constituents,pharmacological activities mechanism and of Xinjiang Bergenia crassifolia is still rarely reported,especially on chemical constituents,purification,isolation and identification.In this article,dried leaves of Bergenia were used as original testing subject,which were extracted and purified by the 70% ethanol reflux method,solvent extraction system,silica column chromatography,polyamide column chromatography,sephadex LH-20 column chromatography,macroporous resin column chromatography,and recrystallization.After going through all the refining process,12 compounds were isolated.These compounds were tested the purity by using thin layer chromatography method and high performance liquid chromatography method.The chemical structures of the 11 compounds were identified by analyzing the results of spectral analysis,comparing with other academic references,etc.As the result,the 12 compounds were thirty-two dioxane(1),β-sitosterol(2),twenty-sixth alkyl ether(3),3-hydroxy myristic acid(4),13-docosenoic acid(5),terephthalic phenol(6),daucosterol(7),bergenin(8),gallic acid(9),propyl gallate(10),11-O-Galloylbergenin(11),and arbutin(12).Among all these compounds,thirty-two dioxane(1),twenty-sixth alkyl ether(3),3-hydroxy myristic acid(4),13-docosenoic acid(5)and propyl gallate(10)were isolated in the plant for the first time.The results of this study not only lay the foundation for research on Xinjiangnational traditional Chinese medicine and development of new drugs,also have the important significance and application value of develop medicinal resources of Xinjiang,inheritance of ethnic folk medicine and development characteristics of traditional Chinese medicine. |
