Objective:Acute renal failure is a serious complication of the anticancer drugcisplatin.The potentialrole of Rgl,a Chinese traditional medicine oncisplatin-induced renal injury isunknown.Pretreatmentwith Rgl ameliorated the cisplatin-induced renal oxidative stress,apoptosis,inflammation andimproved kidney injury and function.Methods:The mice kidney epithelial cell line was cultured and treated with co-administration of cisplatin plus Rgl one group and another group with co-administration of cisplatin plus stem cell.For dose dependent test,cell was cultured for 24 hours with different doses of Cisplatin,Rg1 and Stem cell.Then cell proliferation was examined by cell counting kit CCK-8.The apoptosis and proliferation of mice kidney epithelial cell were determined by DAPI and Fluorescent antibody technique respectively.For understanding the mechanism whether the effect of Rgl via the anti-inflammatory,anti-apoptosis study,or by reducing oxidative stress determined by the expression level of BCL-2,Bax,Bim,caspase 3,caspase-9 were measured by Western blot.Results:For dose-dependent test,low concentration of Rg1 and stem cell stimulated cell proliferation observed by CCK8 and Cell counting assay.Moreover,while the dose within the range of2 ug/ml,4 ug/mlRgland 100%mice stem cell stimulated cell proliferation.the expression level of NF-?B,cleaved caspase-3,andBax is significantly lower in cisplatin treated cell,stimulated by Rg1 than control,but the effect of stem cell was little weaker than Rgl.Conclusions:Rgl is traditional Chinese medicine have been used for cellular healing and restorative properties.Ginsenoside induced cytoprotection was reflected in the increase of cell viability.The molecular mechanism of Rgl attributed by the downregulation of the NK-kB/NO signaling pathway with elevated proportion of Bc12/Bax,and lessened the release of cytochrome C from mitochondria into cytoplasm and then blocked mitochondrial apoptotic cascades.These results suggested that Rg1 may play an important role in the treatment of cisplatin induced nephrotoixicity in the future.
