Study On Thy Chemical Constituents Of Rabdosia Jabdosia

The Rabdosia japonica is Plectranthus genus of Labiatae.It is a perennial herb.According to now research,Labiatae Isodon contain plentiful chemical constituents.In recent years,people have been separated out a variety of monomer compounds from the blue Rabdosia,but complete system research on blue Rabdosia Chemical Components has not yet formed.Thus,this experiment is based on previous studies,further to study the chemical composition of blue Rabdosia,the study tcan provide a basis for further research and development in its medicinal value.This experiment set up 95%ethanol heat reflux extraction method to extract the Rabdosia japonica root and obtained the ethanol extraction.The ethanol extraction was diluted with water and extracted by Petroleum ether,Ethyl acetate,n-Butanol,The combined organic layers was concentrated under vacuum.The extractions was separated by modern means of silica gel column chromatography,reverse phase silica gel column chromatography,high pressure preparation,Sephadex-LH-20 gel column were isolated,purified.The monomeric compounds were authenticated by 1H-NMR,13C-NMR,MS and theirs physicochemical properties.According to its physical and chemical properties,spectral data and TLC chromatography were identified ten compounds.They are the Methyl salicylate(1),?-sitosterol(2),Heptadecan-1-ol(3),Stigmasterol(4),Ursolic acid(5),Oleanolic acid(6),Quercetin(7),Apigenin(8),Daucosterol(9)and Maslinic acid(10).Compound 10 Maslinic acid was first isolated from the Rabdosia japonica.The experiment identified 24 compounds by GC-MS method from the ethanol extraction of the Rabdosia japonica.They were 3-Hexanone,2-Methyl-pentanal,2,4-Dimethyl-l-hexene,3-Hexyl alcohol,2-Methyl-pentadec-l-ene,Furfural,2-Cyclo pentene-1,4-dione,1,2,6-Hexanetriol,2-Methoxy-phenol,2-Methoxybutanedioate,Isobornyl,12-Methyl-trideca-1,5,9,11-tetraene,a-Humulene,4-Isopropylidene-1,2-divinyl-cyclohexane,(+)-?-Selinene,(-)-y-Cadinene,Diisobutyl phthalate,2-Propylc yclohexanol,2,4-Diisopropenyl-1-methyl-1-vinyl-cyclohexane,Phthalic acid dibutyl ester,1-Dodecene,Diisooctyl phthalate,2-24-nonene-1-ol.The Ethyl acetate extract layer and n-Butanol extract layer have good antioxidant activity by antioxidant screening test.The compounds 7 and 10 have excellent antioxidant activity.The IC50 with the compound 7Maslinic acid and compoundlO are 0.60 ?M and 0.48 ?M.The compound 1 Methyl salicylate and compound 6 Oleanolic acid also have some antioxidant activity.
