Objective: To investigate the efficacy of total alkaloids of corydalis saxicola bunting on invasion and migration of human lung cancer A549 cells and its correlation with Vav1/Cdc42.Methods: A549 cells in logarithmic proliferation were treated with 0?g/mL,1.25?g/mL,2.5?g/mL,5?g/mL,10?g/mL,20?g/mL,40?g/mLdosages of total alkaloids of corydalis saxicola bunting.The cytotoxic effect of TAOCSB on A549 cells was evaluated by Methylthiazolyldiphenyl-tetrazolium bromide(MTT)assay.The ability of migration and invasion of A549 cells were analyzed by wound-healing assay and transwell chamber invasion assay,respectively.Next,we tested the effect of TAOCSB on Cdc42?MMP-9?MMP-2 protein by western blotting assay;after using Cdc42 inhibitor with 10?g/mL TAOCSB,the expression quantity of MMP-9?MMP-2 were tested again.Last,we detected the transcriptional level of Cdc42 and Vav1 by real-time PCR.Results: MTT assay revealed that TAOCSB had a little influence on the survival of A549 cells under the dosage of 10?g/mL.Then by using the dosage of 5?g/mL,7.5?g/mL,10?g/mL,TAOCSB could inhibit the migration and invasion of A549 cells in concentration dependent.Moreover,TAOCSB could inhibit MMP-9?MMP-2 and Cdc42 protein.Last,we found TAOCSB could suppress the expression of Cdc42 mRNA and promote the expression of Vav1 mRNA in a dose-dependent manner.Conclusions: TAOCSB could inhibit the invasion and migration of A549 cells in a low-dosage.By detecting the expression of MMP-9?MMP-2 and Cdc42,we suspected that TAOCSB could inhibit the metastasis of A549 cell lines by suppressing Cdc42?MMP-2?MMP-9 protein and specially via targeting Cdc42,but the mechanism of upregulated Vav1 mRNA by TAOCSB remained further study.
