Objective Osteosarcoma was hackneyed malignant tumor with highly malignant biological behaviour in clinic,It was prone to metastasis and prognosis was bad,hypoxia in cell was a important stimulus signal for proliferation of tumour cell,formation of neoformative blood vessel was necessary for migration and metastasis of tumor,the thesis would had a study to expression of HIF-la(anoxic acknowledge signal),COX-2(stimulating factor for forming blood vessel)and VEGFR(effector for forming blood vessel)in osteosarcoma and investigate the effect for biological behaviour.and observed the effect of hypoxemia for a infestation in MG-63 osteosarcoma cellsMethods 73 cases of tissue specimen of osteosarcoma was selected as object of study,and 60 cases of tissue specimen of osteochondroma was selected as control,all tissue specimen were stain in immunohistochemistry with SP ways,results was observed with integration,expressive positive rate and intensity of HIF-1?,COX-2 and VEGFR were detected in osteosarcoma and osteochondroma tissue,expressive diifference of HIF-la,COX-2 and VEGFR were contrasted along osteosarcoma with different clinical stage,different differentiation and different tissue origin,expressive correlation of HIF-la,COX-2 and VEGFR in osteosarcoma were analyzed with spearman correlation test.and cultured MG-63 osteosarcoma cells in hypoxemia(hypoxemia groups)and normoxia condition(normoxia groups),and effect of hypoxemia for a infestation in MG-63 osteosarcoma cells were observed by detecting capability change of cell adhesion,cell migration and cell infestation.Results Mostly expressive intensity of HIF-1?,COX-2 and VEGFR was moderate and strong positive in osteosarcoma tissue,and was weakly positive in osteochondroma tissue,expressive intensity of HIF-1?,COX-2 and VEGFR was higher in osteosarcoma tissue than in osteochondroma tissue(P<0.05),expressive positive rate of HIF-la,COX-2 and VEGFR was 95.8%,93.2%and 94.5%in osteosarcoma tissue,and was 18.3%,30.0%and 31.8%in osteochondroma tissue,expressive positive rate of HIF-la,COX-2 and VEGFR was higher in osteosarcoma tissue than in osteochondroma tissue,expressive positive rate of HIF-la was 92.3%,97.0%and 100%,expressive positive rate of COX-2 was 94.6%,97.0%and 100%,expressive positive rate of VEGFR was 92,3%?94.1%? 100%in I stage,stage and III stage osteosarcoma tissue,there was a significant deviation in expressive positive rate of HIF-la,COX-2 and VEGFR in different stage osteosarcoma tissue(P<0.05),,expressive positive rate of HIF-la was 84.6%,96.8%and 100%,expressive positive rate of COX-2 was 69.2%,100%and 96.3%,expressive positive rate of VEGFR was 76.9%.96.8%and 100%in osteosarcoma tissue with well-differentiated,moderately differentiated and poorly differentiated,there was a significant deviation in expressive positive rate of HIF-1?,COX-2 and VEGFR in different differentiated osteosarcoma tissue(P<0.05),expressive positive rate of HIF-1? was 96.8%,96.5%and 91.6%,expressive positive rate of COX-2 was 93.8%,93.1%and 91.6%,expressive positive rate of VEGFR was 93.8%,96.5%and 91.6%in osteosarcoma tissue with osteoblast origin,chondroblast origin and other origin,there was a significant deviation in expressive positive rate of HIF-1?,COX-2 and VEGFR in different origin osteosarcoma tissue(P<0.05).Correlation test shown there was a positive correlation between expression of HIF-1a and COX-2(?=0.492,P=0.000),between expression of HIF-1? and VEGFR(?=0.364,P=0.001),between expression of COX-2and VEGFR(?=0.416,P=0.000).Cultured MG-63 osteosarcoma cells shown that rate of cell adhesion,cell migration and cell infestation was(106± 12.4)%,(117±9.2)%and(112±15.3)%in normoxia groups and was(184±16.2)%,(206±18.4)%and(174±22.9)%in hypoxemia groups,rate of cell adhesion,cell migration and cell infestation was higher in hypoxemia groups than in normoxia groups,the different was significant in statistics(P<0.05).Conclusions:HIF-1?,COX-2 and VEGFR was overexpression in osteosarcoma tissue,expressive degree was closely related with progression and malignant grade biological behaviour of tumor,HIF-1?,COX-2 and VEGFR participate genesis and development of osteosarcoma by regulating each others.Cultured MG-63 osteosarcoma cells shown that cell infestation capability improve in hypoxemia condition,which hint hypoxemia play a important in migration and infestation of osteosarcoma,and detecting HIF-1?,COX-2 and VEGFR could provid molecular biology reference for pretest biological behaviour and evaluating prognosis in osteosarcoma.
