AIM:As a recently discovered signaling pathway,Hippo signaling transduction pathway is a relatively conservative signaling cascade.By governing cell proliferation and apoptosis,Hippo signaling pathway plays an important role in regulating organ size.The purpose of our study was to explore the expression and clinical significance of hippo signaling transduction pathway in human gastric cancer.METHODS:Tumor tissues(n=60),adjacent non-tumor tissues(n=60)and normal tissues(n=60)were obtained from the same person diagnosed as the primary gastric cancer(GC).70 cases chronic atrophic gastritis tissues with intestinal metaplasia(CAG with IM)were obtained from CAG with IM patients by the method of biopsy from the endoscope.Hippo signaling molecules,including Mstl,Lats1,YAP1,TAZ,TEAD1,Oct4 and CDX2,were explored by the means of RT-PCR(real time quantitative polymerase chain reaction).Proteins expression of Mstl,Lats1,YAP1,TEAD1 and CDX2 were explored by the way of Western blot analysis.RESULTS:Mstl,Latsl and Oct4 mRNA expressions showed an increasing tendency from GC tissues to normal gastric tissues,while the mRNA expressive levels of YAP 1,TAZ and TEAD1 were down-regulated(all P<0.01).Mstl and Latsl protein expression presented a similar trend with their mRNA expression.In addition,YAP1 and TEAD1 protein expression in GC were significantly the highest among those protein expression in the four groups(all P<0.01).CDX2 mRNA and protein expressions in CAG with IM group were higher than that in three other groups(all P<0.01).In GC,the mRNA expressive levels of Mstl,Lats1,Oct4,YAP1,TAZ,TEAD1 and CDX2 had a close connection with lymphatic metastasis and tumor TNM stage(all P<0.01),and was not related to the age of the patients.CONCLUSION:Hippo signaling pathway was activated in human gastric cancer and played a significant part in the emergence,progression and metastasis of gastric cancer.Therefore,operating of Hippo signaling transducting pathway can be a potentially target spot for human gastric cancer treatment.
