ObjectiveIlex asprella(Hook.et Arn.)Champ.Ex Benth was regard as one of the co mmonly used traditional Chinese medicine in Lingnan region.to treat a cold fever,cough,fever and Zimbabwe wound thirst,sore throats,stasis pain,tonsillitis,sore throat,bronchitis,whooping cough,etc.,can also be scattered stasis detumescence,used in the treatment of injuries,etc.Mentor Wu Qingguang found in the clinica1 application of hillock megan have fall hematic fat action,but in the literature th ere is no post to megan cholesterol-lowering medicine research reports.Ingredients like this paper to total saponins as the research focus,through a certain techniqu e for total saponins extraction enrichment extract,by hyperlipidemia rat model,eva luation of total saponins site cholesterol-lowering efficacy,and total saponins of pa rts and component separation and identification,as megan medicine clinical work provides a scientific basis for the treatment of hyperlipidemia.Methods1.Uv-vis spectrophotometric method is adopted to establish the Ilex asprella method for the determination of total saponins content.2.Using the response surface optimization method,by investigating the ethanol concentration,extraction time,solvent dosage,extraction times,establish duty megan total saponins extraction process conditions.3.Application of macroporous resin chromatography technology,by investigati ng the resin type,elution velocity,elution solvent,elution solvent dosage,establish macroporous resin for purification of total root saponin from radix et caulis ilicis asprellae extraction process conditions.3.By establishing the heperlipemia model in Rats,cure it in the total saponin in different dose,finally determine the content of Total Cholesterol Triglyceride,L ow Density Lipoprotein,High Density Lipoprotein,leptin,ADP,IL-6?CPR in bloo d serum.4.isolation to applicate TLC?Sephadex LH-20?ODS?HPLC-ELSD?preHP LC chromatographic technique from ethyl acetate extract.Indentificate compound apply to NMR,MS.Results1.By Design Expert 8.0 software to optimizate influencing factor and arcordi ng to actual condition to regulate parameter.the finally extract way is 56%ethanol,extract time:100min,extract times:2,the dose of ethanol:11 times.2 Total saponins from radix et Ilex asprella purification technology:the applic ation of macroporous resin technology to extract saponin compounds,further purifi cation of enrichment,to determine the best resin for AB-8,3 sample velocity on bv/h,50%ethanol elution agent and dosage of eluent 3 bv column(volume),the dynamic adsorption curve of leakage inspection on samples,reaches 70 ml began t o leak,reached 200 ml adsorption into balance.3.Compared with the hyperlipidemia model group,thetotal saponins from Ilex asprella high,medium and low dose can be significantly reduced in rats serum T C,TG,LDL-c,LEP,CRP and iL-6 levels(P<0.01),compared with the hyperlipi demia model group,total saponins from Ilex asprella,low dose can be significantl y increased in rats serum HDL-c levels(P<0.05),total saponins from Ilex asprell a high dose can be significantly increased in rats serum HDL-c(P<0.01)levels(P<0.01),total saponins from Ilex asprella high doses can significantly increase s erum ADP level in rats(P<0.05).4.There were 5 compounds isolated.Through Physieal spectral analysis identifi ed compounds strueture.2 new sulphur triterpenoid glycosides and first isolated an d one was isolated for the first time in the plant,named asprellcosides A,B,C.f rom gangmei.Two compounds have been reported:19 to WuSu acid,hydrogen R andiali acid B.Conclusion1.Use of hyperlipidemia rat model evaluation proved that megan total saponi ns of fall hematic fat action,megan for hills,new research and development to la y the experimental foundation of clinical utility.2.Separate identification of triterpene compounds for saponins monomer fall h ematic fat action research provides the basis,as well as characteristic constituents from radix et caulis ilicis asprellae as quality testing and standards provide a refer ence value.3.Using response surface optimization software,a macroporous resin chromat ography column enrichment of total saponins from radix et caulis ilicis asprellae e ffective parts,by vacuum concentration method,total saponins extract from radix e t caulis ilicis asprellae using ultraviolet spectrophotometry analysis,to detect the c ontents of total saponins extraction of saponins and optimize the extraction technol ogy of total saponins from radix et caulis ilicis asprellae.
